The epidemiological characteristics of influenza B and the genetic variation of the neuraminidase of influenza B virus in Nanjing in 2016

摘要 目的分析2016年南京市乙型流感流行病学特征及病毒NA基因分子进化特征。方法从中国疾病预防控制中心国家流感监测信息系统导出2016年南京市流感监测数据,应用生物信息学软件进行分析。结果2016年南京市流感监测网络实验室共检测流感样病例咽拭子样本5 626份,检测阳性病例771例,占13.70%。乙型流感282例,占流感病毒分离总数的36.58%。乙型流感病毒感染中,主要以Victoria谱系为主(93.62%);发病年龄以≤15岁人群居多,占58.87%(166/282);发病时间主要集中在3~4月,占71.63%(202/282);9株南京株NA全部分布进化树的Group2分支的两个小分支A/B,同B/Brisbane/60/08NA相比,共发生45I→T、73L→I、120I→V等10处氨基酸位点的替换,重要的酶活性位点及耐药位点均未发生变化。结论乙型流感为南京市2016年流感流行主要的优势毒株之一,春季为高发季节,NA氨基酸位点无特异性位点突变。 Objective To discuss the epidemiological characteristics of influenza B in Nanjing in 2016 and to analyze the genetic variation of neuraminidase(NA)of the virus. Methods The surveillance data of influenza were retrieved from information system in Chinese Center for Disease Control and Prevention,data were analyzed using Bioinformatics software. Results In 2016,a total of 5 626 nasopharyngeal swab samples were collected from cases with influenza-like illness and were tested in Nanjing influenza surveillance network laboratories,771 samples were virus positive with the overall positive rate of 13.70%,282 were positive for influenza B virus,accounting for 36.58% of total positive cases and majority(93.62%)were Victoria lineage strains.More than half(58.87%)of cases were 15 years and younger,and most cases(71.63%)appeared in March and April.Phylogenetic analysis of NA sequences showed that 9 strains of local isolates clustered in two small branches A/B of Group 2.Compared with B/Brisbane/60/08 NA,a total of 10 amino acid substitutions(45 I→T,73 L→I,120 I→V)were observed among 9 strains of local isolates.However,no change was found in the important enzyme activity sites and drug resistance site of NA protein. Conclusions Influenza B virus is one of the dominant influenza strains circulating in Nanjing in 2016.Spring is the peak season of the disease and there is no unique mutation in NA protein.
