摘要
目的探究肥大细胞对肾癌细胞侵袭迁移的影响,并探讨其分子生物学机制。方法 786-O和769-P细胞系均来源于原发性肾透明细胞癌,具有相似的遗传和分子特征。将人肾癌细胞786-O和769-P分别分为对照组和实验组。实验组与对照组分别用肥大细胞条件和正常培养基均培养48 h。用Transwell小室实验观察肾癌细胞迁移和侵袭能力改变;以实时定量-聚合酶链反应法检测转录共激活相关蛋白(YAP)的mRNA水平改变;用免疫印迹法检测肾癌细胞YAP蛋白水平改变。结果 786-O细胞系:对照组和实验组的迁移细胞数分别为127. 00±8. 44,199. 00±6. 57; 769-P细胞系:这2组的迁移细胞数分别为193. 00±10. 69,316. 00±14. 59,组间比较这2个细胞系的迁移细胞数的差异均有统计学意义(均P <0. 01)。786-O细胞系:这2组的侵袭细胞数分别为98. 00±6. 34,208. 00±5. 68; 769-P细胞系:这2组的侵袭细胞数分别为122. 00±4. 84,156. 00±10. 59,组间比较这2个细胞系的侵袭细胞数差异均有统计学意义(P <0. 01,P <0. 05)。786-O细胞系:这2组的YAP与β-actin灰度值比值分别为0. 65±0. 03,1. 10±0. 05; 769-P细胞系:这2组的YAP与β-actin灰度值比值分别为0. 44±0. 04,1. 02±0. 01,组间比较这2个细胞系的灰度值比值差异均有统计学意义(均P <0. 01)。769-P YAP过表达细胞的迁移细胞数和侵袭细胞数,组间比较差异均无统计学意义(均P> 0. 05)。结论肥大细胞通过上调肾癌细胞YAP蛋白,促进肾癌细胞侵袭迁移。
Objective To investigate the effect of mast cell on migration and invasion of renal carcinoma and it’s molecular mechanism. Methods Both 786-O and 769-P cell lines were derived from primary renal clear cell carcinoma with similar genetic and molecular characteristics. Renal carcinoma cell lines 786-O and 769-P were cultured with mast cell conditioned medium as experimental group for 48 h,and cultured with ordinary culture medium as control group for 48 h. The changes of migration and invasion ability were investigated through Transwell experiment. The level of Yes-associated protein( YAP) mRNA was detected by real time quantitative polymerase chain reaction. The protein levels of YAP was tested by Western blot. Results Comparison of the number of migratory cells between experimental group( 199. 00 ± 6. 57)and control group( 127. 00 ± 8. 44) in 786-O,the difference wassignificant( P < 0. 01). Comparison of the number of migratory cells between experimental group( 316. 00 ± 14. 59)and control group( 193. 00 ± 10. 69) in 769-P,the difference was also significant( P < 0. 01). The number of invasive cells between experimental group( 208. 00 ± 5. 68) and control group( 98. 00 ± 6. 34) in 786-O showed significant difference( P < 0. 01). The number of invasive cells between experimental group( 156. 00 ± 10. 59) and control group( 122. 00 ± 4. 84) in 769-P also showed significant difference( P < 0. 05). The ratio of the YAP and β-actin gray scale in control group and experimental group in 786-O were 0. 65 ± 0. 03 and 1. 10 ± 0. 05. Compared the two groups,the difference was significant( P < 0. 01). The ratio of the YAP and β-actin gray scale in control group( 0. 44 ± 0. 04) and experimental group( 1. 02 ± 0. 01) in 769-P also was significantly difference( P < 0. 01). In769-P YAP-overexpressed cell line,comparison between experimental group and control group,the differences of the number of migratory cells and invasive cells were both none-significant( all P > 0. 05).
