摘要
以一株海洋细菌Bacillus sp.ms12为实验对象,在绿色化学思想的指导下设计海洋天然产物提取分离综合性实验教学方案。通过优化菌株发酵条件、优选提取分离纯化方法,使实验时间、菌株发酵量、溶剂消耗、环境污染等显著降低,高效率地分离、纯化出目标产物,引导学生使用核磁共振氢谱鉴定产物结构。该实验有助于促进学生全面深刻地认识天然药物化学学科,使学生深入了解海洋微生物天然产物的科学研究流程,增加学生学习兴趣,提升学生操作技能和科技创新能力,从而达到提高教育和教学质量的目的。
One of comprehensive teaching experiments in extraction, separation and structure elucidation of marine microbial natural products using a marine-derived bacterium, Bacillus sp. ms12 as the experimental subject was designed under the guidance of the thoughts of green chemistry. The experimental time, fermentation volume, solvent consumption and environmental pollution are remarkably reduced by optimizing fermentation conditions and optimal selection of the methods for extraction and separation, which can lead to efficient purification of the target compounds. The structures are elucidated by analysis of the proton nuclear magnetic resonance spectroscopy under the guidance. The experiment helps the students promote the thorough and profound insights into natural medicinal chemistry, deepen the understanding of the procedures of marine microbial natural product chemistry, and improve the interest of study, the operating skills, and the scientific and technological innovation ability, which can improve the quality of education and teaching.
