摘要
为了探明红芽芋试管苗生长所需的适宜光照时间和光照强度,以江西铅山红芽芋试管苗为材料,采用组织培养方法研究不同光照时间和光照强度对其生长发育的影响。结果表明:1)在光照时间为8~16h/d时,随着光照时间的延长,江西铅山红芽芋试管苗的新生芽数、新生根数、新生根长、株高、总叶绿素含量、可溶性蛋白含量和可溶性总糖含量显著增加,脯氨酸含量和MDA含量显著降低;全光照培养可显著降低江西铅山红芽芋试管苗的新生芽数、新生根数、新生根长、株高、总叶绿素含量、可溶性蛋白含量和可溶性总糖含量,但脯氨酸含量和MDA含量显著增加。2)在光照强度为1500~45001X时,高光强可使江西铅山红芽芋试管苗的新生芽数、新生根数、新生根长、株高、总叶绿素含量、SOD活性和POD活性显著增加,质膜透性和丙二醛含量显著降低,但对试管苗根系活力影响不显著。因此,在江西铅山红芽芋试管苗培养温度为(25±1)。C、湿度为70%~80%条件下,其适宜的光照培养时间为16h/d,光照强度为4500lX。
Plantlets of Jiangxiqianshan red bud taro (Colocasia esculenta) were employed to test the effects of various light duration and intensity on the growth of C. esculenta using tissue culture technique. Study results demonstrate that 1) as the light duration prolonged (8-16 h/d), new bud number, new root number, new root length, plantlet height, total chlorophyll content, soluble protein content, and total soluble sugar content of red bud taro significantly increased, while proline content and MDA significantly decreased, but the results were opposite under full light culture conditions; 2) High light intensity (1 500-4 500 lx) significantly enhanced the increase of a number of plantlets" parameters including new bud number, new root number, new root length, plantlet height, total chlorophyll content, SOD and POD activity, distinctly decreased plasma membrane permeability and MDA content, and had no significant effects on plantlet root vigor. Optimum light duration and intensity for red bud taro plantlets were 16 h/d and 4 500 lx respectively with the temperature of (25±1)℃ and humidity of 70%-80%.
