摘要
目的:研究海带多糖硫酸酯WPS、WPS-1、WPS-2和WPS-3对碱性成纤维细胞生长因子(bFGF)诱导的大鼠主动脉血管平滑肌细胞(VSMC)增殖的影响。方法:采用组织贴块法培养VSMC并用免疫组织化学染色法对VSMC进行鉴定;采用bFGF诱导VSMC增殖;以MTT法检测海带多糖硫酸酯对bFGF诱导的VSMC增殖的影响;黄嘌呤/黄嘌呤氧化酶产超氧阴离子自由基(O2-.)体系测定海带多糖硫酸酯对O2-.的清除力。结果:在0.1~1mg/mL质量浓度范围内,WPS、WPS-1和WPS-3对bFGF诱导的VSMC增殖具有抑制作用,其中WPS的增殖抑制性表现显著。WPS-2则对VSMC增殖基本无影响。在O2-.清除力上,WPS-2对O2-.清除率仅为34%,而其他组分自由基清除率均超过70%。结论:WPS、WPS-1和WPS-3对bFGF诱导的VSMC增殖均有显著抑制作用,且抑制作用可能与它们的自由基清除能力相关。
Objective: To explore the inhibitory effect of sulfated polysaccharides from Laminaria japonica on rat vascular smooth muscle cells (VSMCs) induced by basic fibroblast growth factor (bFGF). Methods: Rat VSMCs were obtained from the aorta of adult Sprague Dawley rats by attached explant method and identified by immunohistochemistry assay. The proliferation of VSMCs was induced by bFGF. The inhibitory effects of crude sulfated polysaccharide and its fractions on the proliferation of VSMCs were evaluated by MTT assay. Superoxide anion-scavenging activity of sulfated polysaccharides was determined by xanthine/xanthine oxidase method. Results: At concentrations ranging from 0.1 to 1 mg/mL, sulfated polysaccharides except WPS-2 revealed an obvious inhibitory effect on bFGF-induced proliferation of VSMCs. Similarly, sulfated polysaccharides except WPS-2 showed excellent superoxide anion radical scavenging capability. Conclusion: Sulfated polysaccharides except WPS-2 can dramatically inhibit the proliferation of VSMCs induced by bFGF. The functional mechanism may be related to their antioxidant activities.
