摘要
目的研究穿心莲内酯(andrographolide,Andro)对碱性成纤维细胞生长因子(basicfibroblastgrowthfactor,bF—GF)诱导的血管新生的影响。方法在人原代培养的脐静脉内皮细胞上(humanumbilicalvascularendothelialcell,HUVEC),采用MTF法观察Andro对bFGF诱导HUVEC细胞增殖的影响;采用transwell小室检测Andro对bFGF诱导细胞迁移的影响;采用Matfigel检测Andro对bFGF诱导内皮细胞管腔形成的影响。结果25μmol·L^-1浓度的Andn,可以明显抑制hFGF诱导的HUVEC细胞增殖。1~25μmol·L^-1。不同浓度的Andro均可以抑制bFGF诱导的HUVEC细胞迁移,并呈现明显的剂量依赖性。2.5~25μmol·L^-1不同浓度的Andro均可以抑制bFGF诱导的HUVEC细胞管腔形成,并呈现明显的剂量依赖性。结论Andro可以抑制bFGF诱导的血管新生。
Aim To investigate the inhibition of an- drographolide (Andro) on angiogenesis induced by basic fibroblast growth factor (bFGF). Methods Cell viability was analyzed by 3- (4, 5-dimethylthiazol- 2-yl) 2, 5-diphenyltetrazolium bromide (MTF) assay in human umbilical vascular endothelial cell (HU- VEC). The effects of Andro on bFCF-induced cell mi- gration were detected by Transwell assay. The effects of Andro on bFGF-induced tube formation were ana-lyzed by Matrigel assay. Results Andro (25 txmol ~ L-~) inhibited bFGF-induced cell proliferation. More- over, Andro inhibited bFGF-induced cell migration and tube formation in a concentration-dependent manner. Conclusion Andro can inhibit bFGF-induced angio- genesis in vitro.
