摘要
目的:探讨电针治疗慢性疲劳综合征的作用机制。方法:将96只SD大鼠随机分为空白组、模型组、模针组、侧脑室假注射组(假注射组)、侧脑室注射促肾上腺皮质激素释放因子组(CRF组)、侧脑室注射促肾上腺皮质激素释放因子电针组(CRF电针组)、侧脑室注射促肾上腺皮质激素释放因子拮抗剂组(CRFA组)、侧脑室注射促肾上腺皮质激素释放因子拮抗剂电针组(CRFA电针组),每组12只。除空白组外,其他各组均采用慢性束缚复合强制性冷水游泳复制慢性疲劳模型。假注射组行侧脑室假注射,CRF组、CRF电针组行侧脑室促肾上腺皮质激素释放因子注射,CRFA组、CRFA电针组行侧脑室促肾上腺皮质激素释放因子拮抗剂注射。模针组、CRF电针组、CRFA电针组电针"肾俞""足三里",每天1次,每次20min,治疗3d。治疗后检测各组下丘脑室旁核促肾上腺皮质激素释放激素(CRH)表达、血浆促肾上腺皮质激素(ACTH)及皮质酮(CORT)含量。结果:模型组、假注射组、CRFA组、CRFA电针组下丘脑室旁核CRH表达灰度值较空白组升高(均P<0.05),CRF组、CRF电针组CRH表达灰度值较空白组降低(均P<0.05),模针组、CRF组、CRF电针组CRH表达灰度值较模型组降低(均P<0.05);模型组、假注射组、CRF组、CRF电针组、CRFA组、CRFA电针组血浆ACTH含量较空白组降低(均P<0.05);模型组、假注射组、CRFA组血浆CORT含量较空白组降低(均P<0.05)。结论:电针可使慢性疲劳大鼠下丘脑CRH表达升高,血浆ACTH及CORT含量升高,下丘脑室旁核CRH是电针调整慢性疲劳大鼠下丘脑-垂体-肾上腺轴功能的关键物质。
Objective To explore the mechanism of electroacupuncture on chronic fatigue syndrome. Methods Ninety six SD rats were randomly divided into a blank group, a model group, an electroaeupuncture (EA) group, a sham injection (SI) group, an injection of corticorophin-regulating factor (CRF) group, a CRF injection and EA (CRFEA) group, an injection of CRF antagonist (CRFA) group and a CRFA injection and EA (CRFAEA) group, 12 cases in each one. The chronic fatigue model was made by chronic restraint and mandatory swimming in cold water in all the rats except those in the blank group. The SI group was treated with sham injection in lateral cere bral ventricle. The CRF group and CRFEA group were treated with injection of CRF in lateral cerebral ventricle, while CRFA group and CRFAEA group were treated with injection of CRFA in lateral cerebral ventricle. The EA group, CRFEA group and CRFAEA group were treated with electroacupuncture at "Shenshu" (B1, 23) and "Zu sanli" (ST 36) for 20 minutes, once each day. After treatment of 3 days, the expression of corticotropin release hormone (CRH) in necleus of hypothalamus and the content of adrenocorticotrophic hormone (ACTH) and corti eosterone (CORT) in plasma were detected. Results The gray values of CRH in neeleus of hypothalamus in the model group, SI group, CRFA group and CRFAEA group were significantly higher than those in the blank group(all P〈0.05), while the gray values in CRF group and CRFEA group were significantly lower than those in blank group (both P〈0.05), and the gray values in EA group, CRF group and CRFEA group were significantly lower than those in model group (all P〈0.05). The content of plasma ACTH in model group, SI group, CRF group, CRFEA group, CRFA group and CRFAEA group were all significantly lower than that in the blank group (all P〈 0.05). The content of plasma CORT in the model group, SI group and CRFA group were significantly lower than that in blank group (all P〈0.05)
