摘要
目的:观察益智汤对淀粉样蛋白前体蛋白(APP)695转基因小鼠行为学及脑组织低密度脂蛋白受体相关蛋白(LRP-1)和高糖基化终产物受体(RAGE)表达的影响,探讨益智汤治疗阿尔茨海默病(AD)的作用机制。方法:13月龄APP695转基因小鼠随机分为模型组、益智汤组、同月龄同背景转基因阴性小鼠正常组。益智汤组小鼠每天根据体质量以益智汤(1mL/100g)灌胃,模型组和正常组给予等体积蒸馏水。中药治疗3个月后,使用电迷宫检测各组小鼠学习记忆能力,使用免疫组化和Western Blot技术观察各组小鼠大脑LRP-1和RAGE的表达情况。结果:电迷宫检测发现模型组小鼠学习记忆能力低于正常组(P<0.01),益智汤组小鼠学习记忆能力较模型组小鼠明显改善(P<0.01)。免疫组化和Western Blot结果提示:与正常组相比,模型组小鼠脑组织LRP-1的表达降低,RAGE的表达增加(P<0.01);益智汤治疗90d后,与模型组相比,益智汤组小鼠脑组织各LRP-1的表达增强,RAGE的表达降低(P<0.05)。结论:APP695转基因小鼠脑组织LRP-1和RAGE的表达改变,益智汤通过调整LRP-1和RAGE的表达来改善其学习记忆能力。
Objective: To observe the effect of Yizhi Decoction on the behavior and the expression of LRP-1 and RAGE of APP695 transgenic mice, and to discuss the mechanism of Yizhi Decoction for treating Alzheimer's disease (AD). Methods: The 13-month-old APP695 transgenic mice were randomly divided into model group and test group, the same aged negative transgene mice were normal group. The mice in test group were given intragastrically Yizhi Decoction (lmL/100g), and the model group and normal group were given equal quantity of distilled water. After 3 months the learning and memory abilities of these mice were tested by using ~/-electric maze test, and the expressions of LRP-1 and RAGE in mouse cerebral cortex and hippocampus were observed by applying immunohistochemical and Western blotting assay in all groups. Results: The learning and memory abilities were lower in model group compared with normal group (P〈0.01), while that of Yizhi Decoction group was improved (P〈0.01). The result of immunohistochemical and Western blotting assay indicated that the expressions of LRP-1 were significantly lower and the expressions of RAGE were significantly higher in model group compared with normal group, while the expressions of LRP-1 were significantly higher and the expressions of RAGE were significantly lower in Yizhi Decoction group compared with normal group (P〈0.05). Conclusion: Yizhi Decoction could ameliorate AD syndrome through regulating the expression of LRP-1 and RAGE, which may be one of the mechanisms for treating AD.
