摘要
目的:探讨复方丹参片治疗阿尔茨海默病的作用机制。方法:APP/PS1双转基因小鼠随机分为模型组、复方丹参片组(低、中、高剂量)、多奈哌齐组,同月龄KM小鼠作为正常对照组。给予相应药物治疗2个月后,Western Blot法检测脑组织LRP-1(低密度脂蛋白受体相关蛋白1)、RAGE(晚期糖基化终产物受体)的含量。结果:与正常组比较,模型组小鼠脑组织中RAGE表达显著升高,LRP-1表达显著降低;差异有统计学意义(P<0.05,P<0.01);与模型组比较,各给药组小鼠脑组织RAGE表达明显降低,LRP-1表达明显升高,差异具统计学意义(P<0.05,P<0.01),复方丹参片高剂量组小鼠脑组织中LRP-1表达最高,多奈哌齐组表达较低,且与多奈哌齐组有明显差异(P<0.05);复方丹参片高剂量组小鼠脑组织中RAGE的表达最低,多奈哌齐组表达较高,2组比较无统计学意义(P>0.05)。结论:复方丹参片可能通过调节LRP-1/RAGE的表达水平治疗阿尔茨海默病。
Objective: To investigate the protective mechanism of Fufang Danshen Tablet on Alzheimer's disease. Methods: The APP / PS1 transgenic mice were randomly divided into model group,Fufang Danshen Tablet group( low,middle and high dose groups),Donepezil group,KM mice were used as normal group. After given the corresponding drugs for two months,used the Westen Blot and image analysis method to determine the expressions of LRP- 1 and RAGE in mice's brain tissues. Results: The RAGE expression in brain tissue of model group mice was significantly raised to compare with normal group mice,the LRP- 1 expression was significantly lowered,the differences were statistical significance( P〈0. 05 or P〈0. 01); the The RAGE expression in brain tissue of each medicine group was obviously decreased and the LRP- 1 expression was increased to compare with model group,the differences were statistical significance( P〈0. 05 or P〈0. 01); the LRP- 1 expression of the high dose Fufang Danshen Tablet group was the highest,that was higher than that of Donepezil group( P〈0. 05 or P〈0. 01),the RAGE expression of high dose Danshe Tablet group was the lowest,that of Donepezil group was little higher,the differences had no statistical significance. Conclusion: Fufang Danshen Tablet may ameliorate Alzheimer's disease by regulating LRP- 1 / RAGE expressions.
