摘要
目的:探讨自发性高血压大鼠海马β淀粉样蛋白(Aβ)代谢通路的变化,揭示Aβ代谢通路异常在高血压认知损害中的意义。方法:以SHR和WKY大鼠为研究对象,使用RT-PCR和Western blot方法分别检测SHR及WKY海马RAGE、LRP-1、APP和BACE的mRNA及蛋白质表达水平。结果:与WKY大鼠相比,SHR海马组织RAGEmRNA、BACEmRNA的表达水平明显增高(P<0.05),而APP mRNA表达水平相对降低(P>0.05),LRP-1mRNA表达水平相对增高(P>0.05);与WKY大鼠相比,SHR海马组织RAGE、BACE的表达水平明显增高(P<0.05),而APP和LRP-1表达水平相对增高(P>0.05)。结论:SHR海马RAGE和BACE的mRNA及其蛋白表达水平异常增高,提示Aβ代谢异常可能参与了高血压认知损害的发生机制。
Objective:To explore the changes ofβ-amyloid proteins(Aβ)metabolic pathways in spontaneously hypertensive rats and reveal significance of Aβmetabolic abnormalities in cognitive impairment related to hypertension.Methods:SHR and WKY rats as the research object,using RT-PCR and Western blot method to detect mRNA and protein expression level of RAGE,LRP-1,APP and BACE in SHR and WKY hippocampal.Results:Compared with WKY rats,the expression levels of RAGEmRNA and BACEmRNA in SHR hippocampal tissue increased significantly(P〉0.05),whILe the APP mRNA expression level relativly decrease(P 〉0.05),LRP-1mRNA expression level was relatively higher(P〉0.05);Compared with WKY rats,the expression level of RAGE and BACE in SHR hippocampal tissue signIFIcantly increased(P 〉0.05),whILe the APP and LRP-1expression level was relatively higher(P〈0.05).Conclusion:Abnormal expression of mRNA and protein of RAGE and BACE in hippocampus is associated with SHR cognitive impairment,it is to say that Aβmetabolic abnormalities may be involved in the mechanism of cognitive impairment due to high blood pressure.
