摘要 采用荧光光谱和圆二色谱学分析,研究表没食子儿茶素没食子酸酯(EGCG)对嗜水气单胞菌的作用机制。结果表明,EGCG对嗜水气单胞菌的最小抑制浓度为50μg/ml。光谱学分析结果表明EGCG以嵌插方式结合到细菌DNA的双螺旋碱基对中。 EGCG(epigallocatechin-3-gallate) is the main ingredient of green tea extracts and possesses versatile biological activities including broad spectrum antimicrobial activity.In this experiment,antimicrobial activity of EGCG against Aeromonas hydrophila was investigated.As a result,the minimal inhibitory concentration(MIC) of EGCG was 50 μg/ml.The interaction between EGCG and DNA of Aeromona hydrophila in physiological condition(pH 7.4) was studied by fluorescence and CD spectrophotometer.The result of fluorescence spectroscopy demonstrated that EGCG could bind the phosphate group of DNA and be intercalated into the base pairs in a helix of DNA,which would inhibit cellular functions,and ultimately lead to cell death.
