摘要
拟态弧菌是一种比较罕见的人类和水产动物共患致病菌,目前拟态弧菌毒力的相关研究较少。表没食子儿茶素没食子酸酯(EGCG)是茶叶中多酚类物质最主要的成分,具有良好的抗菌抗病毒性能。测定了EGCG对拟态弧菌的最低抑菌浓度为256μg/mL。用64μg/mL EGCG和256μg/mL EGCG与拟态弧菌共培养24 h后提取毒素粗提液,用毒素粗提液诱导小鼠巨噬细胞RAW264.7。采用流式细胞仪测定细胞凋亡率。结果证实,没有经EGCG孵育的拟态弧菌毒素粗提液致小鼠细胞凋亡率高达75.16%,用64μg/mL EGCG和256μg/mL EGCG孵育后,拟态弧菌毒素粗提液导致小鼠细胞凋亡率仅为9.43%和11.88%。证实EGCG能显著抑制拟态弧菌的毒力。研究结果为EGCG在食品行业中的应用和拟态弧菌的相关研究提供了参考数据。
Vibrio mimicus is a rare pathogen that causes disease in aquatic animals and humans.Little information involving Vibrio mimicus virulence has been revealed.Epigallocatechin-gallate(EGCG),which is the major polyphenolic component of tea,exhibits prominent antibacterial and antivirus activities.In this study,the minimum inhibitory concentration(MIC)of EGCG against Vibrio mimicus was determined to be 256μg/mL.EGCG(64μg/mL and 256μg/mL)and Vibrio mimicus were co-cultured for 24 h.The bacterial supernatants containing toxin were then collected and prepared.Cell apoptosis of mice macrophage RAW 264.7 was induced by the supernatants and detected by using flow cytometry.The results showed that the ratio of cell apoptosis of mice macrophage induced by Vibrio mimicus was 75.16%.However,the toxin supernatants treated with 40μg/mL of EGCG and 200μg/mL of EGCG induced only 9.43%and 11.88%mice macrophage to apoptosis,respectively,indicating that EGCG reduced Vibrio mimicus virulence.The results of this study provide an understanding of Vibrio mimicus and the application of EGCG in food industry.
