血浆HSP70在CUMS诱导抑郁样行为过程中的变化规律 被引量:3

摘要 目的观察慢性不可预计温和应激(chronic unpredictable mild stress,CUMS)诱导抑郁样行为过程中血浆HSP70水平的变化规律,探讨血浆HSP70作为诊断抑郁症的分子标志物的可能性。方法利用CUMS建立大鼠抑郁动物模型,电子秤称量大鼠体质量,糖水偏爱实验、旷场实验评定CUMS模型大鼠行为学的变化,双抗夹心酶联免疫方法检测血浆HSP70水平。结果 (1)在第4~8周,CUMS模型大鼠的糖水偏爱度较对照组显著下降(P<0.05),并呈现逐渐降低的趋势;(2)CUMS模型大鼠的运动行为较对照组呈现先升高后降低的趋势,第6、8周CUMS模型大鼠的旷场实验评分较对照组明显降低(P<0.05);(3)第2、4、6、8周CUMS模型大鼠体质量与对照组相比显著下降(P<0.05);(4)第4、6、8周CUMS模型大鼠血浆HSP70水平较对照组显著升高(P<0.05),并呈现逐渐升高的趋势。结论 CUMS可诱导大鼠产生抑郁样行为,在大鼠抑郁样行为形成过程中,血浆HSP70出现逐渐升高的趋势,提示血浆HSP70具有成为抑郁症诊断生物标志物的可能性。 Objective To observe the change of plasma HSP70 in depression rats induced by chronic unpredictable mild stress(CUMS) and investigate the possibility of plasma HSP70 being used as the diagnostic biomarker for depression.Methods Depression was induced by CUMS,body mass was measured by the electronic animal scale,and sucrose preference test and open-field test were applied to evaluate the behavior of the CUMS rats.The plasma HSP70 level was detected by ELISA.Results(1)Preference sucrose of the CUMS rats decreased significantly from 4 weeks to 8 weeks compared with control groups(P0.05);(2)CUMS rats showed significantly reduced body mass from 2 weeks to 8 weeks compared with the control groups(P0.05);(3)CUMS rats displayed significantly reduced scores of open field test from 6 weeks to 8 weeks compared with the control groups(P0.05);(4)Plasma HSP70 level was significantly higher in CUMS rats than that of the control groups from 4 weeks to 8 weeks(P0.05),and an increasing trend was showed.Conclusion The behavioral changes of rats induced by CUMS are similar to the features of human depression.During the development of rats depression induced by CUMS,plasma HSP70 shows an increasing trend.The results suggest that plasma HSP70 might be the diagnostic biomarker for depression.
