摘要
目的采用高效液相色谱-蒸发光散射法测定养胃颗粒的黄芪甲苷含量。方法色谱柱为Zorbax Extend C18柱(250 mm×4.6 mm5,μm),流动相为乙腈-水(37∶63),流速1.0 mL/min,漂移管温度40℃,N2气体压强3.5 Bar。结果黄芪甲苷质量浓度在0.048～0.48 mg/mL范围内与峰面积线性关系良好(r=0.999 1),平均加样回收率为97.7%,RSD=1.25%(n=6)。结论高效液相色谱-蒸发光散射法简便、准确、重复性好,可用于养胃颗粒的黄芪甲苷含量测定。
Objective To determine the content of astragaloside IV in Yangwei Granules by HPLC-ELSD.Methods HPLC-ELSD was used in the quantitative analysis by using Zorbax Extend C18 chromatography column（250 mm × 4.6 mm,5 μm） and acetonitrile-water（37 ∶ 63） as the mobile phase.The flow rate was 1.0 mL/min.The drift tube temperature was 40 ℃.The gas pressure was 3.5 Bar.Results The linear range of astragaloside IV was 0.048-0.48 mg/mL（r = 0.999 1）.The average recovery rate of astragaloside IV was 97.7% with RSD of 1.25%（n = 6）.Conclusion The method is simple and accurate with good reproducibility,which can be adopted for the determination of astragaloside IV in Yangwei Granules.
