摘要
[目的]建立一种简单、灵敏的测定面粉中过氧化苯甲酰含量的方法。[方法]在中性或弱酸性溶液中,氨存在条件下亚硫酸盐可与苯胺生成强荧光物质,而过氧化苯甲酰对该荧光物质具有抑制作用。基于该原理,对反应溶剂、pH值、发射波长、苯胺用量、亚硫酸钠用量及反应时间等进行优化,建立测定面粉中过氧化苯甲酰含量的荧光光度法。[结果]过氧化苯甲酰在8×10-8~1×10-6g/ml范围内与其荧光强度呈线性关系。优化后的反应条件为,无水乙醇为溶剂,pH值为6.56,发射波长为340nm,苯胺用量为2.0ml,亚硫酸钠用量为2.5ml,反应时间为30min。该方法的检出限为0.03μg/ml,相对标准偏差RSD为1.4%。[结论]该研究建立了测定面粉中过氧化苯甲酰含量的新方法。
[ Objective] The research aimed to establish a simple and sensitive content determination method for benzoyl peroxide in flour. [ Method ] In neutral and weak acidic solution, sulfite could react with phenylamine to generate fluorescent substance when there was ammonia and benzoyl peroxide had inhibition effect on this fluorescent substance. Based on this rationale, the reaction solvent, pH value, emission wavelength, phenylamine dosage, sulfite dosage and reaction time were optimized for establishing fluorimetry for determining benzoyl peroxide content in flour. [ Result] In the range of 8×10^-8~1×10^-6g/ml, the benzoyl peroxide content showed linear relationship with its fluores- cence intensity. The optimized reaction conditions were as follows: the absolute ethanol was used as solvent, the pH value was 6.56, the emission wavelength was 340 nm, the phenylamine dosage and sulfite dosage were 2.0 and 2.5 ml and the reaction time was 30 min. The detection limit and relative standard deviation (RSD) of this method were 0.03 μg/ml and 1.4% resp. [ Conclusion] This research established a new method of determining benzoyl peroxide content in flour.
