SUMO-1在实验性大鼠肝纤维化形成过程中的作用 被引量:4

Role of small ubiquitin-related modifier-1 in the pathogenesis of hepatic fibrosis in rats

摘要 目的:探讨小泛素相关修饰物(SUMO-1)在肝纤维化形成中的作用机制.方法:将90只♂SD大鼠分为2组:模型组(n=50)和对照组(n=40),对模型组给予40% CCl4 3μL/g皮下注射,分别于造模后2、4、6、8、10、12wk取肝组织,同时给予对照组等量的生理盐水皮下注射,且分别在与模型组相同的时间处死动物.采用病理检查观察肝纤维化形成的变化,RT-PCR检测SUMO-1 mRNA的表达,Western blot检测SUMO-1蛋白的表达,将各周检测的数据进行分析,相互比较,并将各周数据分别与对照组进行比较分析.结果:随着肝纤维化至肝硬化的形成,SUMO-1mRNA在2、4、6、8、10、12wk的表达逐渐增强(0.725±0.017、0.786±0.018、0.803±0.023、0.831±0.020、0.863±0.016、0.892±0.008,P<0.01).Western blot显示SUMO-1的蛋白表达在2、4、6、8、10、12wk也是同时逐渐增强(0.810±0.059、0.873±0.049、0.923±0.055、0.959±0.032、0.988±0.011、0.998±0.004,P<0.01).结论:SUMO-1基因及蛋白表达在肝纤维化形成过程中逐渐增强,说明其在肝纤维化形成过程中具有重要的意义. AIM:To observe the changes in the expression of small ubiquitin-related modifier-1 (SUMO-1) during the formation of hepatic fibrosis in rats and to investigate the role of SUMO-1 in the pathogenesis of hepatic fibrosis. METHODS: Ninety male Sprague-Dawley rats were divided into two groups: model group and control group. The model group was subcutane- ously injected with 40% carbon tetrachloride at a dose of 0.3 mL/100 g of body weight, while the control group was given equivalent volume of normal saline. Liver tissue samples were taken at weeks 2, 4, 6, 8, 10 and 12 after carbon tetra- chloride injection. Hepatic fibrosis was patho- logically evaluated. The expression of SUMO-1 mRNA and protein in liver tissue was detected by RT-PCR and Western blot, respectively.RESULTS: During the formation of hepatic fi- brosis, the expression of SUMO-1 mRNA was gradually up-regulated from week 2 to 12 (0.725 ± 0.017, 0.786 ± 0.018, 0.803 ± 0.023, 0.831 ± 0.020, 0.863 ± 0.016 and 0.892 ± 0.008, respectively; P 0.01). Similarly, SUMO-1 protein expression was also gradually up-regulated from week 2 to12 (0.810 ± 0.059, 0.873 ± 0.049, 0.923 ± 0.055, 0.959 ± 0.032, 0.988 ± 0.011 and 0.998 ± 0.004, respec- tively; P 0.01). CONCLUSION: The expression of both SUMO-1 mRNA and protein is gradually up-regulated during the formation of hepatic fibrosis, suggest- ing an important role of SUMO-1 in the patho- genesis of hepatic fibrosis.
