摘要
目的研究肝癌传代细胞、肝癌标本及癌旁肝组织中SUMO-1基因的表达及意义。方法采用RT-PCR、Westernblot方法在mRNA及蛋白水平检测肝癌传代细胞、肝癌标本及癌旁肝组织中SUMO-1基因的表达。结果在mRNA及蛋白水平,SUMO-1基因在肝癌传代细胞SMMC-7721、Hep3B、HepG2及肝癌标本中均明显高表达,而癌旁肝组织中SUMO-1基因明显低表达,肝癌组织和癌旁肝组织中SUMO-1表达差异有统计学意义(P<0.001)。结论SUMO-1基因在肝癌中高表达,SUMO-1可能为肝癌诊断和治疗中的一个靶点。
Objective To investigate the expression and clinical significance of SUMO-1 in SMMC-7721, HepG2, Hep3B,hepatocellular carcinomas(HCC) and liver tissues surrounding HCC. Methods The mRNA and protein of SUMO-1 in SMMC-7721, HepG2, Hep3B, HCCs and liver tissue surrounding HCCs were examined by means of RT-PCR and Western blot. Results The expression level of SUMO-1 mRNA and protein elevated significantly in SMMC-7721, HepG2, Hep3B and hepatocellular carcinoma tissues. Although the quantities of SUMO- 1 mRNA and protein varied in liver tissues surrounding HCC,there was a significant difference between hepatocellular carcinoma tissues and their surrounding liver tissues (P〈0. 001). Conclusion The expression of SUMO-1 in HCCs shows marked contrast enhancement relative to the liver tissues surrounding HCC. SUMO-1 may be an interest- ing target in the diagnosis and treatment of HCC.
