摘要
目的探讨实验性脑出血大鼠脑组织中缺氧诱导因子(HIF-1α)在脑出血后不同时间点的动态演变过程及促红细胞生成素(EPO)对其影响。方法应用自体血脑内注射法建立脑出血大鼠模型并随机分为假手术组、脑出血非干预组及脑出血EPO干预组,应用免疫组化方法检测脑出血后不同时间及组别血肿周边脑组织中HIF-1α蛋白表达情况。结果大鼠脑出血后HIF-1α蛋白阳性表达率在72 h内随时间进行性增加,并于出血后72 h达到高峰,此后HIF-1α蛋白表达的阳性率逐渐下降,脑出血EPO干预组较脑出血组各时间点HIF-1α蛋白阳性率明显减少,差异有统计学意义(P<0.01)。结论脑出血后HIF-1α表达存在一定变化规律,EPO能显著减少HIF-1α表达。
Objective To investigate the dynamic changes of HIF-1α in intracerebral hemorrhage(ICH) model rats and the influence of rhEPO on it.Methods The model of ICH in rats was established by intracerebral injection of autogenous blood,then the models of rat with ICH were divided into three groups,sham-operation group,model group and intervention group.The expressions of HIF-1α in cerebral tissue around the hemorrhagic focus and the normal cerebral tissue at different time points and different groups after ICH were detected by SP immunohistochemistry. Results The expression of the HIF-1 was gradually increased within 72h after intracerebral hemorrhage and reached peak at 72h. After that the expression of the HIF-1 was gradually decreased, and there were significant differences in different time points ( P 〈 0. 01 ). The positive expression rate of the HIF-1 after intervened by rhEPO was significantly decreased at different time points, as compared with that in model group ( P 〈 0. 01 ). Conclusion There are certain change rules in the expression of HIF-1a after intracerebral hemorrhage. And rhEPO can significantly reduce the expression of HIF-1α.
