补骨脂对乳腺癌骨痛大鼠痛行为及肿瘤生长的影响 被引量:16

Effects of Fructus Psoraleae on Tumor Growth and Pain Behaviors in Rats with Bone Pain Induced by Breast Cancer

摘要 目的研究补骨脂对大鼠乳腺癌骨痛模型痛行为、骨破坏及肿瘤生长情况的影响。方法将大鼠乳腺癌细胞MRMT-1株接种在雌性SD大鼠左侧胫骨骨髓腔内制成乳腺癌骨痛模型,观察补骨脂(水煎剂,造模次日开始给药,剂量为生药0.5g.100g体重-1.d-1)的作用,以唑来膦酸作为阳性对照药。造模20d后观察Von Frey纤维刺激的50%缩足阈(50%PWT)、双足负重差异;第21日取材,测定子宫系数和肿瘤体积;用左胫骨放射评分、骨密度(BMD)和骨矿物质含量(BMC)评价骨破坏程度。结果补骨脂可使乳腺癌骨转移大鼠50%PWT提高,双足负重差异下降,肿瘤体积减小,放射评分减低,BMD和BMC升高。结论补肾壮骨中药补骨脂在大鼠乳腺癌骨痛模型上能减轻癌性疼痛,其机制可能与抑制肿瘤生长、减轻骨破坏有关。 Objective: To study effects of Fructus Psoraleae on pain behaviors, bone destruction and tumor growth in a rat model of cancer pain induced by Bone Metastasis from breast cancer. Methods: Sprague - dawley rats receiving intra - tibial injections of syngeneic MRMT - 1 rat mammary gland carcinoma cells developed behavioural signs indicative of pain. Fructus Psoraleae extract was given to model rats, Zoledronic acid for injection as contrast. On day 20 after injection with MRMT - 1, Behavior of 50% Paw withdrawal threshold(50% PWT) and the difference of weight bearing between hind paws were observed in all groups. All rats were killed by the day 21, volume of tumor was measured , the uterus coefficient was calculated. The development of the bone structural damage to the bone was monitored by radiological analysis, quantitative measurement of bone mineral density(BMD) and bone mineral capacity(BMC). Results: After the treatment of Fructus Psoraleae, 50% PWT of fructus psoraleae group was increased and weight bearing difference between hind paws was reduced, compared with model. BMD and BMC was enhanced, radiographs showed less damage in fructus psoraleae group than model. Conclusion: Herba epimedii can alleviate pain of bone cancer and inhibit tumor growth and bone destruction induced by cancer.
