摘要
目的观察补骨脂提取物对体外培养新生大鼠颅骨成骨细胞增殖、分化的影响。方法在新生大鼠颅骨成骨细胞体系中加入不同浓度的补骨脂提取物,检测细胞内碱性磷酸酶(ALP)含量,并用MTT法检测细胞增殖。结果补骨脂乙酸乙酯提取物和乙醇提取物能促进大鼠颅骨成骨细胞ALP活性及其增殖,最佳浓度为2.0×10-3mg/mL。结论补骨脂乙酸乙酯提取物和乙醇提取物均可促进成骨细胞分化、增殖。
Objective To investigate the effect of Fructus Psoraleae on the proliferation and differentiation of cultured osteoblast isolated from neonatal rat＇ s calvarium. Methods Calvarium osteoblasts of new born SD rat were cultured and exposed to different concentrations of Fructus Psoraleae extract, then the content of alkaline phosphatase（ALP） in the cell was measured and the cell proliferation was detected by tetrazoline colorimetry（MTY）. Results Ethyl acetate extract and alcohol extract from Fructus Psoraleae can improve the activity of ALP and the proliferation of rat＇ s cultured osteoblasts, the optimum concentration was 2.0 × 10^-3 mg/mL. Conclusion Ethyl acetate extract and alcohol extract from Fructus Psoraleae can stimulate the differentiation and proliferation of osteoblasts in vitro.
