含纤维素酶细菌的筛选及其纤维素酶生物学性质的研究 被引量:9

Screening of Bacteria Producing Cellulase and Investigation of Biological Characteristics of the Enzyme

摘要 以上海崇明养殖场堆肥为筛选材料,通过刚果红染色鉴定,获得一株高活力纤维素酶生产细菌。研究表明该菌为芽孢杆菌,适宜在pH值7.0~8.0、37~42℃条件下生长。该菌的CMC酶活力的最适pH值8.0,酶反应的最适温度50℃,酶活力最大时可达68.6U/ml。Ca^2+、Cu^2+、Co^2+是纤维素酶的促进剂,高浓度的Cd^2+和Pb^2+是纤维素酶的抑制剂,并且该纤维素酶对乙醇热耐受能力较强。 A Bacillus strain was isolated from muck of Chongming which had high endo-β-glucanase producing ability proved by the Congo red dying method on culture plates. Optimal growing conditions of the strain were 37-42℃, pH value 7.0-8.0. It showed that the optimal pH value and temperature of cellulase activity (CMCase) were pH 8.0 and 50℃, and the maximal activity of CMCase is 68. 6 U/ml. In addition, Ca^2+ , Cu^2 +and Co^2+were the activators, Cd^2+. and Pb^2+ were the inhibitors of the enzyme. The cellulase was insensitive to ethanol.
