镉致胎鼠骨骼发育异常的形态学观察 被引量:6

摘要 目的观察镉对胎鼠骨骼发育的影响。方法将孕6 d的孕鼠随机分成4组,一次性腹腔注射氯化镉,剂量依次为0、1、2、4 mg Cd/kg体重,处死孕20 d的孕鼠,取出胎鼠,检查胎鼠的发育情况,并对部分胎鼠行茜素红染色。结果不同组之间的胎鼠体重、颅臀长、头长存在着显著性差异,并随着体内染镉剂量的加大而减小;三种不同剂量染镉组的胎鼠骨骼发育明显落后于对照组,表现为未骨化或骨化范围明显小于对照组;露脑、脊柱裂、卷尾、脊柱侧弯、肋骨缺失、顶骨缺失和胸骨缺失等发育异常明显多于对照组。结论镉可以导致胎鼠骨骼发育明显迟缓和骨骼畸形。 Objective To observe cadmium-induced dysplasia of skeleton in rat embryos. Methods Pregnant rats of 6 embryo day were divided into 4 groups. Animals of different groups were treated by intraperitoneal injection with a single dose of O, 1, 2 and 4 mg Cd/kg body weight respectively. They were killed at the 20th day of gestation, and their fetus were taken out and checked. The parts of fetus were observed development of skeleton by alizarin red staining. Results The body weight, crown rump length, head length of fetal rats were significant difference between different groups, and decreased with the increase of cadmium dose. In three different cad- mium treatment groups the development of fetal skeleton was more retardant than those in the control group. The mainly performance of skeleton development retardation was that the parts of skeleton were unossification or the scope of ossification was less than that in the control group. The number of dysplasia fetusin in the cadmium treatment groups was more than that in the control group. The mainly dysplasia were exencephaly, spina bifida, coiled tail, scoliosis, rib absence, sternum absence, and so on. Conclusion Cadmium can induce that the skeleton development of fetal rat were delayed significantly and malformation.
