摘要
目的:为防治铅中毒提供实验室研究资料,探讨铅染毒时铅对未成年小鼠骨髓造血干/祖细胞和外周血的毒性作用。方法:60只昆明系小鼠随机分成4组,分别以0mg Pb2+/L、164mg Pb2+/L、546mg Pb2+/L、1638mg Pb2+/L浓度的醋酸铅饮用水对小鼠染毒,于染毒的第10天取小鼠股骨骨髓细胞作体外培养,观察各组红系祖细胞成集落数(BFU-E)和粒系祖细胞成集落数(CFU-GM)的变化并进行外周血测定。结果:醋酸铅可明显抑制骨髓BFU-E和CFU-GM的生长,集落数与对照组相比差异显著(P<0·05),且随醋酸铅浓度的增加,骨髓BFU-E抑制作用增强(相关系数为rs=-0·972,P=0·000),而外周血Hb、WBC和PLT与对照组比较差异无显著性(P<0·05)。结论:骨髓造血祖细胞对铅较为敏感,铅染毒小鼠外周血变化不明显时,已引起骨髓BFU-E和CFU-GM的生长障碍。
Objective: To investigate the toxic effect of acetic acid lead on haematopoietic stem/progenitor cells and peripheral blood in the immature mice of lead poisoning. Methods: 60 mice were divided into four groups and poisoned by acetic acid lead (0, 164 mg, 546 mg, 1 638 mg Pb^2+/L) . BFU - E, CFU - GM, Hb, PLT and WBC were observed after 10 days. Results: The number of BFU - E and CFU - GM in Pb group was significant lower than that in the control group (P 〈 0. 05) . The number of BFU - E was significantly deceased with the increase of Pb concentrations ( rs = - 0. 972, P = 0. 000), while there was no significant difference in Hb, WBC and PLT (P 〉 0. 05) after exposed to Pb for 10 days. Conclusion: The haematopoietic stem/progenitor cells are more sensitive to lead. The reduction of BFU - E and CFU - GM was observed before that of Hb, WBC and PLT.
