摘要
为研究促进排铅功能动物实验适宜模型条件 ,利用三水醋酸铅作为造模物质 ,使用清洁级昆明种健康小鼠 2 0 9只 ,其中雄性小鼠 179只 ,雌性 30只 ,饮水 (16 4mgPb2 + L、5 4 6mgPb2 + L、16 38mgPb2 + L)或饲料喂饲 (2 2 4mg kg、72 8mg kg、2 2 4 0mg kg)给予小鼠染毒 ,分别观察 10d和 2 0d后测定肝、脑、股骨和血中的铅含量。观察小鼠性别、染毒方式、染毒剂量、造模时间对促进排铅功能小鼠模型建立的影响。结果表明小鼠性别对铅负荷小鼠模型无影响。 (16 4mgPb2 + L、5 4 6mgPb2 + L、16 38mgPb2 + L三水醋酸铅水溶液饮水和 2 2 4mg kg、72 8mg kg、2 2 4 0mg kg三水醋酸铅饲料喂饲 ,均可建立稳定的铅负荷小鼠模型。 5 4 6mgPb2 + L三水醋酸铅水溶液饮水染毒 10d即可达到小鼠模型成立的效果 ,该方法可以作为建立铅负荷小鼠模型的方法。
In order to establish a lead poisoning mouse model for evaluating the lead expelling function of a health food,a total of 209 KunMing mice (male 179, female 30) and lead acetate (PbAC) as a poisoning agent were used for study. The animals were randomly divided into 7 groups .Experimental lead overload was produced by drinking water (164 mg Pb 2+ /L、546 mg Pb 2+ /L、1 638 mg Pb 2+ /L) and feeding diet (224 mg/kg, 728 mg/kg, 2 240 mg/kg). At the 10th day and 20th day, the concentrations of lead in liver, brain, femur and blood were measured. The results showed that sex had no effect on the loading of lead, both female and male mice could be used for experiment. The mouse model could be established by drinking water containing PbAC164mg Pb 2+ /L、546 mg Pb 2+ /L、1 638 mg Pb 2+ /L,or feeding diet containing PbAC 224 mg/kg, 728 \{mg/kg,\} 2 240 mg/kg. The water containing PbAC 546 mgPb/L could produce stable mouse model in 10 days. This method can be used for establishing a lead poisoning mouse model.
