摘要
目的:探讨Fas/FasL在小儿皮肤血管瘤不同时期的表达情况及其与血管瘤退化的可能关系。方法:采用免疫组化SP法检测不同时期小儿皮肤血管瘤和瘤体周围正常皮肤组织中Fas,FasL及增殖细胞核抗原(PCNA)的表达;用末端脱氧核苷酸基转移酶介导的脱氧尿苷三磷酸(dUTP)-生物素颈端标记法(TUNEL)检测凋亡细胞。结果:Fas抗原阳性表达率在增生期、退化期和退化完成期分别为33.3%、87.5%和18.7%。退化期的阳性表达高于增生期和退化完成期(P〈0.05)。FasL阳性表达率在增生期、退化期和退化完成期分别为22.2%、75.0%和25,0%,退化期的阳性表达高于增生期和退化完成期(P〈0.05)。PCNA阳性表达率在增生期、退化期和退化完成期分别为83.3%、37,5%和25.0%。增生期组与退化期和退化完成期组比较差异有统计学意义(P〈0.05)。增生期、退化期和退化完成期血管瘤组凋亡指数(AI)分别为1.7778±0.7535、23.3563±14.5164和1.4375±0.6043。退化期组与增生期及退化完成期组比较差异有统计学意义(P〈0.01)。结论:Fas及FasL共同参与了小儿皮肤血管瘤增生与退化的全过程,其机制可能与Fas/FasL介导和调控血管瘤内皮细胞凋亡有关。
Objective To study the expression of Fas/FasL in different stages of infantile dermal hemangiomas and the relationship between the expression of Fas/FasL and the regression of hemangiomas. Methods Fas/FasL and the PCNA expression in hemangiomas and normal skin tissues were detected by SP, and the apoptotic cells in the different stages of infantile dermal hemangiomas were detected by TUNEL assay. Results The positive rates of Fas expression in proliferating, involuting and involuted phases were 33.3%, 87. 5% and 18.7%, respectively, and the positive rates of FasL expression were 22. 2%, 75.0% and 25.0%. The expression rate of Fas and FasL in involuting phase was higher than that in proliferating and involuted phase ( P 〈0. 05) and control group ( P 〈0. 01 ). The PCNA expression rate is 83.3% in the proliferating period, 37.5% in involuting period and 25.0% in involuted period. The expression rate of PCNA in proliferating phase was higher than that in involuting and involuted phase ( P 〈 0. 05 ). Apoptotic cells were seldom observed in proliferating hemangiomas( AI, 1. 7778± 0. 7535 ) and involuted hemangiomas( AI, 1. 4375 ± 0. 6043 ), but significantly increased in involuting phase specimens( AI, 23. 3563 ± 14. 5164). The AI in involuting phase group was higher than that in proliferating phase group and involuted phase group ( P 〈 0. 01 ). Conclusions Expressions of Fas/FasL are involved in the converting of hemangiomas from proliferating phase into involuting phase. The activation of Fas and FasL may induce apoptosis of endothelial cells, thus lead to the regression of hemangioma.
