摘要
目的探讨CD59基因抗He-Ne激光诱导凋亡的作用。方法将小鼠随机分成两组,小鼠背部脾区皮下分别接种HeLa细胞(Ⅰ组)、转CD59基因HeLa细胞(Ⅱ组),接种24 h后采用24 mW的He-Ne激光器照射小鼠脾区(瘤区),然后采用免疫组化方法检测凋亡相关基因Fas、B细胞淋巴瘤-白血病基因2(bcl-2)、细胞核因子-κB(NF-κB)的表达。结果Ⅰ组瘤块质量及bcl-2、NF-κB表达明显低于Ⅱ组,两组比较差异有显著性(t=9.169,Z=-2.328、-2.367,P<0.05)。Ⅱ组Fas的表达与Ⅰ组比较无显著差异(Z=-0.643,P>0.05)。结论CD59基因可促进肿瘤细胞的生长及bcl-2、NF-κB的表达,具有抗He-Ne激光诱导凋亡的作用。
Objective To study the effect of CD59 gene on apoptosis ot HeLa cell mduced by He-Ne Laser. HeLa cells were injected subcutaneously into the back of the mice in group Ⅰ , and HeLa cells transfected with CD59-gene into the back of the mice in group Ⅱ. After 24 h of the iniection, the mice received He-Ne laser radiation. Expressions of Fas, bcl-2 and NF-κB in tumor tissue were demonstrated via immunohistochemistry technique. Results Compared with group Ⅱ , tumor weight and the expressions of bcl-2 and NF-κB in group Ⅰ were significantly lower (t= 9. 169;Z= -2. 328,- 2. 367;P〈0.05), while difference in the Fas expression between the two groups was not significant (Z=-0.643, P〉0, 05), Conclusion CD59 gene may promote the expressions of bcl-2 and NF-κB, suggesting its anti-apoptosis effect,
