摘要
对工程蝇蛆体内活性物质及体外抗肿瘤作用进行了研究。选用SephadexG-50初步分离蝇蛆体内活性物质;采用MTT法测定K562细胞的生长抑制率;通过倒置显微镜观察K562细胞的形态变化。实验结果表明:峰1组分半致死浓度IC50为100μg/mL,峰2组分半致死浓度IC50为80μg/mL。随所添加活性物质浓度的增大,肿瘤细胞受抑制作用增强;活性物质作用于K562细胞前后其细胞形态发生明显变化,受抑制细胞由一个完整细胞变为多个凋亡小体,因此可以说明工程蝇蛆体内活性物质在体外对K562细胞具有抑制生长的作用。
This thesis focuses on the research of anti-tumor function of effective ingredients in musca domestica. Sephadex G-50 is selected to separate effective ingredients in musca domestica tentatively; MTr is adopted to measure the density of half death. The tumor cell is observed the change of its form under the inversion microscopes. IC50 of the component of first peak is about 100μg/mL, and IC50 of the component of second peak is about 80 μg/mL. With the rising of the density, the two samples all strengthen the suppressing rate of tumor cell. Before and after the effective ingredients acts on K562 cell, the obvious change takes place in its cell form. The cell apoptosis from intact cell. So the effective ingredients in musca domestica have suppressing function of growing on K562 cell in vitro.
