摘要
目的探讨枸杞多糖联合自杀基因tk/GCV系统对移植性小鼠肝癌的治疗效果。方法实验分为模型组、枸杞多糖组、单纯自杀基因系统治疗的tk/GCV组、自杀基因系统联合枸杞多糖治疗的联合组;把小鼠肝癌细胞H22的tk+和tk-细胞按1∶4比例混合,按2×106个细胞/只接种昆明小鼠腋下皮下组织内,次日随机分组;第2天开始中药治疗15d,长出肿瘤后tk/GCV系统治疗11d,进行疗效观察。结果从肿瘤体积看,联合组生长抑制程度最大,第10天起与模型组比较差异明显,差异随治疗时间增加而逐渐增大且有统计学意义,第14天抑瘤率达40.9%(P=0.018<0.05)。而tk/GCV组在第12天、枸杞多糖组第10 ̄12天也有明显抑瘤作用(P<0.05),但第14天起与模型组比较差异减少,差异无统计学意义。结论枸杞多糖联合自杀基因系统能抑制小鼠肝癌移植瘤生长,减少瘤块体积。枸杞多糖能提高自杀基因系统对小鼠移植瘤的杀伤力。
[ Objective] To investigate the curative effect of lycium barbarum polysaccharides(LBP) combining with HSV tk/GCV system on ,nurine transplanted hepatocarcinoma. [Methods] H22/tk and H22 wild type cells were ,nixed in the proportion of 1: 4. Then the mixed cells were inoculated subcutaneously into Kunming mice. The tumor-bearing mice were divided into four groups randomly: tumor model control group, GCV treated group, LBP treated group and GCV/LBP combination group, and the experiment was performed. The LBP treated group and the combination group had been treated with LBP from the second day on for 15 days, while the GCV treated group and the combination group had been treated with GCV when the tumor had been detectable. The tumor inhibitory effect was observed by n,easuring tumor growth sizes. When the experiment ended, tumor weights were measured. [ Resuits] The resuh of experiment in vivo indicated that the tumor growth in the GCV/LBP combination group was significantly inhibited from 10th day on, compared with the control group, and the difference became greater and greater with time. The inhibitory rate was 40.9% (n =15, P =0.018〈0.05) on 14th day. Inhibitory tendency was shown in GCV treated group on 12th day and LBP treated group on 10-12th day (P 〈0.05). But the differences were getting smaller from 14th on with no statistical significance. [Conclusion] The effect of therapy combining LBP with HSV-tk/GCV system on routine transplanted hepartocarcinoma is better than the therapy of HSV-tk/GCV system alone. LBP has the possibility of enhancing the sensibility of tk^+/tk^- mixed cells to GCV.
