逍遥散对应激大鼠海马突触体内PKC活性及Ca^(2+)浓度的影响 被引量:7

摘要 目的:探讨逍遥散对慢性心理应激大鼠海马突触体内Ca2+浓度及蛋白激酶C(PKC)活性的影响,进一步研究其对心理应激损伤学习记忆保护作用的机制。方法:Wistar大鼠随机分为五组,采用慢性多相性应激模型,以Fura-2负载及荧光分光光度计检测大鼠海马突触体内Ca2+浓度,采用琼脂糖凝胶电泳法检测突触体内PKC活性。结果:两个模型组大鼠海马突触体内Ca2+浓度及PKC活性均有上升(P<0.01),而逍遥散组与模型组比较,突触体内Ca2+浓度及PKC活性则明显回落(P<0.05或P<0.01)。结论:慢性多相性应激可升高大鼠海马突触体内Ca2+浓度及PKC活性,逍遥散具有对抗应激升高大鼠海马突触体内Ca2+浓度及PKC活性的作用。 Objective: To observe the influence of Xiaoyao powder on the concentration of Ca^2+ and PKC activity in hippoeampal synaptosome of multi stress model rat. Methods: Wistar rats were randomly allocated to five groups. All the rats except those of normal control were replicated to chronic multi-stress models. After the administration, the hippocampal homogenate was taken and loaded by Fura-2 ,the con centration of Ca^2+ were detected by fluorirnetry and the PKC activity by agarose-gel electrophoresis. Resuits: Compared with normal control group, the concentration of Ca^2+ and PKC activity in hippocampal synaptosome of both model groups were increased ( P 〈0.01) . Compared with model groups, both group of Xiaoyao powder were decreased ( P 〈0.05 or P 〈0.01), Conclusion: These observation suggests multi-stress model can increase the concentration of Ca^2+ and PKC activity in hippoeampal synaptosome of rats, and Xiaoyao powder can counteract such changes .
