加味保元汤对束缚应激大鼠海马CA3区神经元损伤的影响 被引量:5

Research on the Effect of Modified Baoyuantang on the Damage of Restrained Stress Rats' Neurons in Hippocampal CA3 Area

摘要 为了研究加味保元汤对束缚应激大鼠海马CA3区神经元损伤的影响,将50只SD大鼠随机等分为正常对照组、模型组、加味保元汤高剂量组、加味保元汤中剂量组和加味保元汤低剂量组。模型组、加味保元汤高剂量组、加味保元汤中剂量组和加味保元汤低剂量组大鼠每日束缚1次,连续3周。加味保元汤高剂量组、加味保元汤中剂量组和加味保元汤低剂量组束缚前分别给予大鼠300 mg/kg体重、200 mg/kg体重和100 mg/kg体重加味保元汤水提取物灌胃。采用苏木素-伊红染色,光镜下观察各组海马CA3区神经元的变化。结果表明,与正常对照组比较,模型组海马CA3区神经元细胞数减少,排列紊乱,多数细胞边界不清,核膜消失,核质空泡样变,细胞核固缩,加味保元汤中剂量组未见上述明显改变。实验结果提示加味保元汤对束缚应激大鼠海马CA3区神经元损伤具有保护作用。 The objective of this research is to study the effect of Modified Baoyuantang on the damage of restrained stress rats' neurons in hippocampal CA3 area. The experiment is conducted by randomly and equally divides 50 rats into 1. Control group; 2. Stressed group; 3. Stressed group fed with high concentration of Modified Baoyuantang; 4. Stressed group fed with median concentration of Modified Baoyuantang; 5. Stressed group fed with low concentration of Modified Baoyuantang. Rats in group 2 to 5 were stressed once per day for 3 continuous weeks. Rats in group 3 to 5 were fed with the extractive of Modified Baoyuantang according to their body weight. The concentration of the extractive differs across groups. For group 3, 4, 5 the concentration of the extractive is 300 mg per kilogram of body weight, 200 mg per kilogram of body weight, 100 mg per kilogram of body weight respectively. Used the light microscope to observe the hematoxylin and eosin stained neurons. Compared to group 1, pyramidal neurons for rats in group 2 decreased in amount and placed in a messy manner. Blurred boundary, vanished caryotheca and pyknosis were observed in most neurons. The abovementioned changes were found not significant in group 4. The result of the experiment suggested that Modified Baoyuantang has protective effect on the damage of restrained stress rats' neurons in hippocampal CA3 area.
