逍遥散对应激大鼠海马神经元内Ca^(2+)浓度及NR_1 mRNA表达的影响 被引量:4

Study of XiaoYao Powder Influences the Expression of Ca^(2+) and NR_1 mRNA in Hippocampus of Chronic-stress Rats

摘要 目的:研究逍遥散对慢性应激大鼠海马神经元内Ca2+浓度及NR1 mRNA表达的影响,探讨逍遥散对应激损伤学习记忆保护的作用机制。方法:采用慢性多相性应激大鼠模型,以Fura-2负载及荧光分光光度计检测大鼠海马突触体内Ca2+浓度,RT-PCR法检测海马神经元内NR1 mRNA表达的变化。结果:模型组大鼠海马突触体内Ca2+浓度及NR1 mRNA表达均明显上升(P<0.01),而与模型组比较,逍遥散组大鼠突触体内Ca2+浓度及NR1 mRNA表达明显回落(P<0.05,P<0.01)。结论:抑制慢性应激所导致的大鼠海马神经元内NR1 mRNA的过度表达,减少神经细胞膜上NR的数量,从而减轻慢性应激状态下海马神经元内Ca2+超载所致的细胞毒性损伤。这可能是逍遥散抗应激损伤的关键机制。 Objective: To observe that XiaoYaoSan(XYS) influences the concentration of hippocampal synaptic Ca2+ and the expression of NR1mRNA in hippocampus of chronic-stress rats.To study the protective mechanisms of XYS to the function about learn and remembrance of chronic-stress rats.Methods: chronic stress rats were as researching object.The concentration of hippocampal synaptic Ca2+ were detected in fluorimetry,and the expression of NR1mRNA in hippocampus were detected in RT-PCR.Results: Compared with normal group,both the concentration of hippocampal synaptic Ca2+ and the expression of NR1mRNA in hippocampus of model group increased obviously(P〈0.01);Compared with model group,both the concentration of hippocampal synaptic Ca2+ and the expression of NR1mRNA in hippocampus of XYS group decreased obviously(P〈0.05;P〈0.01).Conclusion: Restraining the expression of NR1mRNA in hippocampus of chronic-stress rats,lessening the NR of hippcampal neuron,and relieving the Ca2+ overload of hippcampal neuron of chronic stress rats,it is the possible mechanism that XYS resists in chronic-stress injury.
