摘要 目的 探讨急性缺氧条件下 4 DMAP对红细胞生成高铁血红蛋白的效应特点。方法 新鲜兔血离心洗涤后 ,置于特制的 90 %N2 +5 %O2 +5 %CO2 混合气体缺氧罐内孵育 ,以复制急性缺氧红细胞模型。 4 DMAP与缺氧红细胞作用一定时间后 ,采用分光光度法于波长 63 5nm测定高铁血红蛋白浓度。结果 急性缺氧对红细胞的血红蛋白含量无明显影响。当反应体系中 4 DMAP浓度为 6 49× 10 - 7、6 49× 10 - 6 、6 49× 10 - 5、6 49× 10 - 4和 6 49× 10 - 3mol L时 ,缺氧红细胞生成高铁血红蛋白的能力比常氧红细胞分别提高了 2 2 0 %、2 7 0 %、5 1 44 %、3 14 %和 5 0 5 %。升高反应温度和延长缺氧时间均明显提高 4 DMAP药理学效应。结论 缺氧增强 4 Objective To characterize the effect of 4 DMAP on hypoxic erythrocytes in ferrihemoglobin(MHb) formation Methods The experimental erythrocytes were isolated from fresh rabbit blood and incubated in the mixed gases (90% N 2 + 5% O 2 + 5% CO 2) for the preparation of the hypoxic erythrocyte model Ferrihemoglobin concentration (% of control) after the reaction of 4 DMAP and erythrocytes was determined by spectrophotography at the wave length of 635 nm Results There was no difference in hemoglobin content between hypoxia groups and controls It was demonstrated that the ability of ferrihemoglobin formation induced by 4 DMAP at the concentrations of 6 49×10 -7 , 6 49×10 -6 , 6 49×10 -5 , 6 49×10 -4 and 6 49×10 -3 mol/L increased by about 22 0%, 27 0%, 51 44%, 3 14% and 5 05% in hypoxic erythrocytes, respectively Both the increases in reaction temperature and the elongation of hypoxia period significantly enhanced the pharmacological effect of 4 DMAP on hypoxic erythrocytes Conclusion Hypoxia can enhance the effect of 4 DMAP on hypoxic erythrocytes in ferrihemoglobin formation
