低温复合氰中毒对家兔肝肾功能的影响及4-DMAP的解毒效果观察

摘要 目的探讨低温暴露复合氰化钠中毒对家兔肝肾功能的影响及4-二甲氨基吡啶(4-DMAP)的解毒效果。方法30只家兔随机分为6组,20℃组、2℃组、-20℃组、2℃氰中毒组、-20℃氰中毒组、2℃氰中毒治疗组,每组5只。动物经腹腔注射麻醉后行左侧颈总动脉插管术,按分组分别置于20℃室温、2℃和-20℃实验冰柜中。中毒组于置冰柜后60 min时腹腔注射氰化钠2.0 mg/kg;治疗组于置冰柜后60 min时腹腔注射氰化钠2.0 mg/kg,同时肌肉注射4-DMAP 3.2 mg/kg。各组在置于实验温度后0、30、60、90、120 min时间点经颈动脉插管采血并分离血浆,采用生化方法测定肝功能指标,包括门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、碱性磷酸酶(ALP)、L-谷氨酰基转移酶(GGT),以及肾功能指标尿素(BU)、肌酐(CREA)、尿酸(UA)的含量。结果低温暴露和低温复合氰中毒时,血清AST、ALT、BU、CREA、UA含量均有明显升高,复合损伤时升高尤为明显,且致伤时间越长,变化越显著。给予4-DMAP干预后,AST、ALT、BU、CREA的含量较未干预组有所下降。结论低温暴露和氰化钠中毒对兔肝肾功能的影响有联合效应,4-DMAP能部分改善此作用。 Objective To investigate the effects of cold exposure and sodium cyanide intoxication on rabbit hepatic and renal function and anti-cyanide effect of 4-DMAP. Methods 30 rabbits were randomly divided into 6 groups, group 20 ℃ ,2 ℃, -20 ℃, cyanide poison- ing under 2 ℃, -20 ℃ and anti-cyanide with 4-DMAP under 2 ℃ ,5 rats in each group. The rabbits accepted left carotid artery intubation after intraperitoneal injection anesthesia,which are arranged in 20 ℃ ,2 ℃ and -20 ℃ experimental freezer by groups. The cyanide poison- ing group were injected with NaCN at the dose of 2.0 mg/kg after 60 min. The treatment group received intraperitoneal injection of sodium cy- anide ( 2.0 mg/kg ) and intramuscular injection of 4-DMAP ( 3.2 mg/kg ) after 60 min. The blood samples were collected at time points of 0,30,60,90,120 rain ,respectively. The biochemical methods were used for the determination of liver function index including the content of AST,ALT, ALP,CCT and renal function index of urea(BU) ,CREA, UA. Results The content of AST, ALT, BU, CREA and UA were in- creased under cold exposure and sodium cyanide intoxication especially combined injury. While the changes of AST, ALT, BU and CREA con- tent were improved after 4-DMAP intervention. Conclusion Hypothermal exposure and sodium cyanide intoxication have combined effect on the rabbit liver and renal function. 4-DMAP can improve the function.
