摘要
目的探讨血清中MRE11、TPO、Th 17Treg、IL-17、VEGF在儿童再生障碍性贫血免疫发病的诊断价值及治疗意义。方法采用回顾性研究方法,收集2011年8月到2018年2月在北京市大兴区人民医院诊治的再生障碍性贫血患儿42例作为观察组,同期留取42名儿童保健科体检者外周血作为对照组。采集所有小儿的外周空腹静脉血,采用双抗体一步夹心法酶联免疫吸附试验定量检测法检测MRE11、TPO浓度,采用双抗体两步夹心酶联免疫吸附法检测IL-17、VEGF含量,采用qRT-PCR法检测Th 17TregRORΥ1、Foxp3表达水平并进行相关性分析。结果观察组的TPO、IL-17含量高于对照组,MRE11、VEGF含量低于对照组,对比差异有统计学意义(P<0.05)。观察组的Th 17Treg细胞中的RORΥ1表达水平高于对照组,Foxp3表达水平低于对照组,对比差异有统计学意义(P<0.05)。在观察组患儿中,直线相关性分析显示Th 17Treg细胞中的RORΥ1表达水平与TPO、IL-17、血小板计数、巨核细胞数呈正相关性(P<0.05),与MRE11、VEGF含量呈负相关性(P<0.05);Foxp3表达水平水平与TPO、IL-17、血小板计数、巨核细胞数呈负相关性(P<0.05),与MRE11、VEGF含量呈正相关性(P<0.05)。结论血清中MRE11、TPO、Th 17Treg、IL-17、VEGF在儿童再生障碍性贫血中呈现高表达情况,可互相影响,共同参与该病的免疫发病。
Objective To investigate the diagnostic values and therapeutic significance of serum MRE11,TPO,Th17 Treg,IL-17 and VEGF in children with aplastic anemia.Methods Used a retrospective study,from August 2011 to February 2018,42 children with aplastic anemia diagnosed were selected as he observation group,and the 42 children′s health checkups in the peripheral blood were selected as the control group in the same period.The peripheral fasting venous blood were collected from all children,the concentration of MRE11 and TPO was detected by double antibody onestep sandwich enzyme-linked immunosorbent assay(ELISA),the levels of IL-17 and VEGF were detected by two-step sandwich enzyme-linked immunosorbent assay,the expression levels of Th 17 TregRORΥ1 and Foxp3 were detected by qRT-PCR and were given correlation analysis.Results The levels of TPO and IL-17 in the observation group were higher than those in the control group,and the contents of MRE11 and VEGF were lower than those in the control group,compared the difference were statistically significantly(P<0.05).The expression level of RORΥ1 in Th 17 Treg cells in the observation group were higher than that in the control group,and the expression level of Foxp3 were lower than that in the control group,compared the difference were statistically significant(P<0.05).In the observation group,linear correlation analysis showed that the expression level of RORΥ1 in Th 17 Treg cells were positively correlated with TPO,IL-17,platelet count and megakaryocyte count(P<0.05),and were negatively correlated with MRE11 and VEGF levels.Sex(P<0.05);Foxp3 expression level were negatively correlated with TPO,IL-17,platelet count and megakaryocyte count(P<0.05),and were positively correlated with MRE11 and VEGF content(P<0.05).Conclusion MRE11,TPO,Th 17 Treg,IL-17 and VEGF in serum show high expression in children with aplastic anemia,which can affect each other and participate in the immune pathogenesis of the disease.
