摘要
目的 :探讨γ 干扰素眼内应用时对眼组织的毒性反应 ,为γ 干扰素的临床应用提供实验依据。方法 :前房注药组 :大耳白兔 15只 ,分为 5组 ,向前房内注入γ 干扰素 0 .2ml,浓度分别为 5 0万IU、2 5万IU、12 .5万IU和 6 .2 5万IU ,对照组注射平衡盐液 0 .2ml。晶状体囊袋内注药组 :大耳白兔12只 ,分为 4组 ,右眼经透明角膜切口行超声乳化晶状体摘除术并植入人工晶状体 ,向囊袋内注入γ 干扰素 0 .2ml,浓度分别为 5 0万IU、2 5万IU及 12 .5万IU。对照组同时行手术 ,术中囊袋内注入平衡盐液 0 .2ml。结果 :前房注药组 :5 0万IU组用药后 1~ 3d前房反应与对照组相比差异有显著性(P <0 .0 5 ) ,2 5万IU组用药后第 1天前房反应与对照组相比差异有显著性 (P <0 .0 5 ) ,其余各组与对照组相比差异无显著性 (P >0 .0 5 )。所有实验组兔眼角膜透明 ,眼底未见异常 ,眼压与对照组相比差异无显著性。病理切片检查 :5 0万IU用量组有一只眼见虹膜少量炎症细胞浸润 ,其余实验组兔角膜、虹膜及视网膜均未见异常反应。晶状体囊袋内注药组 :5 0万IU用量组术后 1~ 3d前房反应经单因素方差分析 ,与对照组相比差异有显著性 (P <0 .0 5 ) ,其余各组与对照组相比差异无显著性 (P >0 .0 5 )。病理切片检查所有实验组角膜。
Objective:To investigate the toxicity of various doses of gamma-interferon(γ-IFN) administered intraocularly in the rabbit.Methods:①γ-IFN injection into the anterior chamber:fifteen rabbits were randomly divided into 5 groups. Six eyes in each group received anterior chamber injections of 500000 IU,250000 IU,125000 IU or 62500 IU of γ-IFN reconstituted in a 0.2 ml balanced salt solution,while 6 eyes in the control group received an anterior chamber injection consisting of the same volume of a balanced salt solution.②γ-IFN injection into the lens capsule:twelve rabbits were randomly divided into 4 groups. Each group received phacoemulsification with intraocular lenses implantation through a corneal incision and doses of 500000 IU,250000 IU or 125000 IU of γ-IFN reconstituted in a 0.2 ml balanced salt solution injected into the lens capsule before finishing the surgery. The control group underwent the same surgery and a 0.2 ml balanced salt solution was injected into the lens capsule.Results:①γ-IFN injection into the anterior chamber:injection of 500000 IU or 250000 IU of γ-IFN elicited more severe anterior chamber inflammation than in the control group(P<0.05). No abnormality was found in the cornea,iris,retina or intraocular pressure for all 30 eyes. Histopathological study 10 days after injection showed a slight inflammation from cell infiltration in one eye that received 500000 IU of γ-IFN.②γ-IFN injection into the lens capsule:doses of 500000 IU induced more severe anterior chamber inflammation than for the control eyes(P<0.05) but the effect of doses of 250000 IU of γ-IFN or 125000 IU of γ-IFN were no different than for the control eyes(P>0.05). Histopathological study 3 months after injection did not show any pathologically toxic effects.Conclusion:γ-IFN appears to be safe and is well tolerated for doses up to 125000 IU.
