摘要 目的研究环孢素A(CsA)在不同情况下,对兔晶状体上皮细胞(RLECs)超微结构和增殖的影响。方法用CsA分别处理正常生长的RLECs和经过H2O2损伤的RLECs后,用透射电镜观察RLECs超微结构变化;噻唑蓝(MTT)法测定RLECs增殖能力改变。结果CsA浓度≥10μmol/L时,明显损伤正常RLECs的超微结构和增殖;当浓度<40μmol/L时,能部分抑制H2O2对RLECs超微结构损伤和增殖抑制。与对照组比较,差异有显著意义(P<0.01)。结论CsA对不同情况下的兔晶状体上皮细胞的超微结构和增殖具有不同的作用。 Objective To investigate the effect of cyclosporine A ( CsA) on the ultrastructure and proliferation of lens epithelial cells(LECs). Methods Rabbit lens epithelial cells were cultured in vitro,and oxidative damage model was induced by expositing the cultured LECs to 0. 1 mmol/L H2O2 for 2 hours. Different concentrations of CsA were added for 2 hours or 30 min in normal cultured LECs and oxidative damaged cells respectively. Transmission electron microscope and MTT assay were separately used to observe the changes of LECs ultrastructure and proliferation. Results 0. 1 mmol/L H2O2-treated RLECs showed nucleus chromatin condensation and substructure disappearance in part, and intact mitochondrial structure could be seen in 10 μmol/L CsA-pretreated RLECs. 20 μmol/L CsA-treated RLECs showed mitochondria bubble and cristae disappearance in LECs and deceased the proliferation of LECs. However, CsA less than 40 μmol/L showed a partly protection on H2O2-induced damages of cell ultrastructure and inhibited H2O2-induced effect on proliferation of LECs. Conclusion CsA has dual effects on rabbit lens epithelial cells in different conditions.
