摘要
目的:探讨白细胞介素1β(IL1β)反义寡核苷酸脂质体(ASODN)对后发性白内障形成的影响。方法:30只兔行白内障囊外摘除术。单纯白内障囊外摘除术为对照组,手术时前房注入单纯脂质体为liposomes组,注入ASODN为ASODN组。定期抽房水,采用酶联免役吸附实验检测房水IL1β的含量,定期进行临床观察。术后3个月用光镜观察晶体后囊的形态学变化。结果:术后17d房水IL1β的浓度,ASODN组明显低于对照组,差异有显著意义(P<0.05)。术后3个月ASODN组晶体后囊透明度优于对照组(P<0.05)。对照组晶体后囊前覆盖一层晶体上皮细胞,ASODN组晶体后囊前无明显晶体上皮细胞增殖。结论:IL1βASODN可能能够有效抑制后发性白内障的发生。
Objective: To investigate the effect of IL-1β antisense oligodeoxynucleotide(ASODN) liposomes in prevention of posterior capsular opacification. Methods. Extracapsular cataract extraction was performed on 30 rabbit eyes which were divided into three groups:control, liposome and ASODN group. The first control group were injected PBS in the operation, the liposome group were injected liposome, and the third were injected ASODN liposomes of IL-1β into the capsular bag during cataract extraction. The aqueous humors were drawn regularly and determined the concentration of IL-1β by enzyme-linked immunosorbent assay with the Quantikine ELISA Kit system. Clinical observation was carried out regularly. Three months later, the biomicroscopic photograph of posterior capsule and the pathological changes of posterior lens capsule were observed by microscopy. Results: IL-1β level in aqueous humors of ASODN group during 1-7 days after operation were significantly lower than those in the control and liposome groups. Three months after the operation, the transparency of the lens posterior capsule in the drug experimental group was superior to that in the control group (P〈0.05). The lens posterior capsule was covered by a layer of lens epithelium in the control group, while the lens epithelium could hardly been observed on the lens posterior capsule in the ASODN group. Conclusion: Antisense oligodeoxynucleotid liposomes of IL-1β may effectively prevent posterior capsular opacification.
