摘要
目的 观察参麦对环孢霉素A所致肾小管细胞毒性损伤的影响。方法 利用体外培养的环孢霉素A中毒模型 ,测定细胞培养上清液LDH活性及参麦对环孢霉素A中毒后肾小管细胞增殖活性的影响。结果 环孢霉素A中毒细胞胞浆内出现空泡变性 ,细胞增殖活性明显降低 ,上清液LDH活性明显升高。参麦可明显减轻环孢霉素抑制细胞增殖的作用 ,上清液中LDH活性明显降低。结论 参麦可对抗环孢霉素A的肾毒性作用 ,促进损伤的肾小管上皮细胞增生。
Objective:To investigate the effects of Shen-Mai on cyclosporin-induced toxicity to cultured renal tubular epithelial cells in vitro.Methods:The toxicity model of cultured renal tubular cell strain induced by CsA was used. The proliferative activity of tubular cell was measured with the method of MTT. LDH level in culture supernatant was detected.Results:Histology revealed vacuolization in tubular cell, and that the prolifative activity of tubular cells decreased significantly and the activity of LDH increased in the cells of CsA-induced cytotoxicity in vitro, while these changes became less conspicuous in cells treated with Shen-Mai.Conclusion:The findings indicated that cyclosporin A might contribute to renal toxicity, which could be inhibited by Shen-Mai.
