摘要
目的探讨环胞霉素A(CsA)诱导的急性肾损伤大鼠模型中肾脏热休克蛋白70(HSP70)的分布及表达规律。方法150只大鼠随机分为对照组A和实验组B〔CsA2mg/(kg.d)注射〕,实验组C〔CsA5mg/(kg.d)注射〕,将对照组和实验组分别于注射后1、3、6、12、24h各取10只大鼠剖腹取肾,光镜、电镜下观察肾脏的病理变化,通过免疫组化法观察HSP70的表达情况。结果HSP70在肾皮质肾小球周围近曲小管上皮细胞胞浆内分布增加,其表达量在注射后1h已经升高,3h达到高峰,24h仍高于正常对照组。同一时相点C组较B组HSP70的表达量差异显著(P<0.01)。结论一次小剂量注射CsA即可引起急性肾损伤,在损伤的早期HSP70的表达亦显著增加,提示肾毒性药物早期即可引起肾小管上皮细胞的应激反应,可能与细胞的自我保护机制启动有关。
Objective To illustrate the expressional distribution of heat shock protein 70 (HSP70)in the kidney of acute renal injury rats caused by cyclosporin A. Methods 150 rats were randomly divided into three groups:group A (normal group), group B[2mg/(kg·d) cyclosporin A treated), group C [5mg/(kg·d) cyclosporin A treated). The variations of renal pathology were observed at different time points by light scope and electron microscope. Simultaneously, the expression of HSP70 in kidney was evaluated by immunohistochemistry. Results HSP70 distributed in epithelial cells of renal tubular in acute injury rats. The expression of HSP70 increased markedly at 1 hour after injection, peaked after 3 hours and lasted for 24 hours. The expression of HSP70 in group C was higher than group B( P 〈 0.01 ). Conclusion Once small dosage of cyclosporin A could cause acute renal injury in rats. It suggests that the increase of HSP 70 expression may be the result of cell self-protection mechanism.
