摘要
采用有机溶剂法从仙人掌中提取皂甙化合物,用硅胶柱层析与SephadexG-75柱分别进行分离纯化,得到纯品.并建立了仙人掌总皂甙的测定方法,以浓硫酸氧化仙人掌皂甙化合物,采用紫外光分光光度法,在最大吸收波长276nm处找出最佳的显色条件为:浓硫酸用量为2.0mL,反应温度60℃,反应时间80min,相关系数r=0.9898,变异系数RSD=1.16,样品平均回收率为100.2%.结果表明:该法操作简便可靠.
The saponins were extracted from Opuntia with organic solvent and purified by silica gel column and Sephadex G-75 gel chromatography. And a method was developed for determination of the content of total saponins in Opuntia. The saponins were oxygenated with concentrated sulfuric acid and determined by spectrophotometry at 276 nm. The optimal color conditions were as follows: the quantitative of concentrated sulfuric acid was 2.0 mL, the reaction temperature was 60℃, and the reaction time was 80 minutes. Relative standard deviation and the average recovery were 1.16% and 90.08 respectively. The method is simple and reliable.
