摘要
以3-氢头孢二苯甲酯为原料,经苯酚水解、固定化青霉素酰化酶PGA-450裂解制得头孢母校(7-ANCA),并优化了工艺条件,确定去4位二苯甲基最佳反应温度范围为60~65℃,去7位苯乙酰基的最佳温度范围为30~32℃,最佳pH 9.0。在此条件下进行了400批次实验,转化率和收率均无明显降低。得到的产物经HPLC测定了含量,并用红外、元素分析进行了结构表征。
7-amino-3-nor-eephalosporanic acid(7-ANCA)was prepared from 3-norcephalosporin via hydrolysis by phenol and enzyme cleavage.It was found that the optimal temperature of hydrolysis was 60~65℃,the optimal temperature of enzyme cleavage reaction was 30~32℃,and the optimal pH was 9.0.Under the reaction conditions,a batch experiment was carried out for 400 times,and the conver- sion and yield did not decrease evidently.The product was assayed by HPLC and characterized by IR and element analysis.
