摘要
目的 :建立测定血浆中扑痫酮 (primidone ,PMD)、苯巴比妥 (phenobarbital,PBB)、苯妥英 (phenytoin ,PHT)、卡马西平(carbamazepine ,CBM )浓度的HPLC法。方法 :以C18反相柱 (15 0mm× 4 .6mm ,5 μm)为色谱柱 ,流动相为甲醇 水 (5 0∶5 0 ) ,流速为 0 .8mL·min-1,检测波长 :2 0 8nm ,以乙酸乙酯为提取剂。结果 :PMD、PBB、PHT、CBM高、中、低 3种浓度的平均回收率分别为 10 4 .7% ,97.8% ,10 1.9% ,10 2 .7% ,日内、日间RSD均低于 5 % ,四药线性范围均为 :1.0~ 80 .0mg·L-1。结论 :方法灵敏、准确、简单 ,可用于PMD、PBB、PHT和CBM的临床血浓监测。
OBJECTIVE To establish an HPLC method for quantitative determination of primidone,phenobarbital,phenytoin and carbamazepine in blood plasma.METHODS Blood plasma samples were extracted with hexane to remove lipid and interfering substances;PMD,PBB,PHT and CBM then were extracted with ethyl acetate.The residues dissoved were analyzed with a reversed phase HPLC column ( 4.6 mm×150 mm,5 μm);Mobile phase was MeOH-H_2O(50∶50) 0.8 mL·min -1 ;UV detection was at 208 nm.RESULTS The average recoveries for PMD,PBB,PHT and CBM were 104.7% , 97.8% , 101.9% and 102.7% ,respectively.The within-day and between-day relative standard deviations for the four compounds were all less than 5%.The calibration curves for the four compounds had good linearity, r = 0.9999 ,within a concentration range of 1.0 ~ 80.0 mg·L -1 .CONCLUSIONS The method provides a sensitive,accurate,precise and reliable analytical procedure for clinical monitoring of plasma PMD,PBB,PHT and CBM.
