摘要
目的建立1种能同时检测5种常用抗癫痫药的血药浓度的HPLC法。方法以Shim-pack VP-ODS(150mm×4.6 mm,5μm)为分析柱,流动相为乙腈-甲醇-0.05 mol.L-1磷酸盐缓冲液(30∶28∶42,v/v),检测波长240 nm。以SPE预处理血浆样品后进样分析。结果5种药物在线性浓度范围内线性关系良好,平均回收率均>90.0%,日间、日间RSD<10.0%。结论建立的方法准确、快速,可同时测定5种抗癫痫药的血药浓度。
Objective To establish an HPLC method for assay of 5 anti-epilepsy drugs in the plasma. Methods The chromatographic column was shim-pack VP-ODS （150 min× 4.6 mm, 5μm）, with a solution of acetonitrile -methanol-0.05 mol · L^-1 phosphate buffer solution （30 ： 28 ： 42, v/v）. The column temperature was 30 ℃ and the UV detector was 240 nm. After SPE pretreatment, the supernatant was injected as sample. Results The 5 drugs presented a good linear correlation within the linear range, the average recovery exceeded 90%, while the intra-day and inter-day RSD was less than 10%. Conclusion The method is accurate and rapid, and can be used for the assay of simultaneous determination of 5 drugs in the plasma.
