摘要
用高效液相色谱法建立了氯硝安定血浓度测定法。本法以茶碱为内标,用二氯甲烷提取,氮气吹干,流动相溶解进行测定。色谱条件:国产YWG—SiO2柱(4.6mm×250mm),紫外检测波长254nm,灵敏度0.020AUFS,流动相为正己烷—无水乙醇(52∶48)。血药浓度在10.0~200.0μg/L范围内线性良好,日内相对标准偏差为2.0%~4.9%,日间相对标准偏差为2.3%~5.2%,最低检测限1.0μg/L。本法灵敏度较高,用血量少,操作简便,已应用于临床治疗药物监测。
A simple, sensitive and rapid normal phase HPLC method for the determination of clonazepam (CZP) in human serum is described, using YWG-SiO2 column (4. 6mm×250mm) and UV detector at 254nm. The mobile phase was hexane-ethanol (52 : 48), the flow rate was 0. 9ml/min. The linear range of drug concentration in serum was 10. 0~200.0μg/L (r=0. 9993) and detection limit was 1.0μg/L. The recovery rate of CZP was 99. 2%~101. 3%, the RSD of within-day and between-days were 2. 0%~4. 9% and 2. 3% ~5. 2%, respectively. The method was already used in clinical practices.
