摘要
目的建立测定血清中苯巴比妥(phenobarbital,PBB)、苯妥英(phenytoin,PHT)、卡马西平(carbamazepine,CBM)浓度的HPLC法。方法色谱柱为ShimadzuODS柱(150mm×4.6mm,5μm),保护柱为C18柱(1cm×4.6mm,5μm),流动相为乙腈∶甲醇∶水(2∶50∶50,内含1%醋酸和0.25%三乙胺,pH5.0),流速为0.8mL·min-1,检测波长:230nm,室温检测,以乙腈为提取剂。结果PBB、PHT、CBM高、中、低3种浓度的平均回收率分别为98.2%,101.7%,102.1%,日内、日间RSD均低于5%,三种药物在1.0~50.0mg·L-1浓度范围之间线性良好。对119例患者血清浓度监测,临床效果满意。结论方法灵敏、准确、快速,可用于PBB、PHT和CBM的临床血药浓度监测。
Objective To establish an HPLC method for quantitative d et ermination of phenobarbital,phenytoin and carbamazepine in blood serum.M ethods Blood serum samples were extracted with hexane to remove lipid and interfering substances;PBB,PHT and CBM then were extracted with acetonitrile.Th e residues dissolved were analyzed with a reversed phase HPLC column ( 4.6 mm × 150 mm,5 μm); Mobile phase was acetonitrile∶MeOH∶H2O(2∶50∶50,1%Acet ic acid and 0.25%Triethylamine,pH=5.0) 0.8mL·min -1;UV detection was at 230nm.Results The average recoveries for PBB,PHT and CBM were 9 8.2%,101.7%,and 102.1%,respectively.The within-day and between-day relative standard deviations for the three compounds were all less than 5%.The calibrati on curves for the three compounds had good linearity,r= 0.999 9,within a con centration range of 1.0~50.0 mg·L -1.The clinical application was good. Conclusions The method provides a sensitive,accurate,precise an d reliable analytical procedure for clinical monitoring of serum PBB,PHT and CBM .
