摘要
目的:分析消渴汤对糖尿病大鼠肝脏糖脂代谢及炎症反应影响,为临床患者治疗提供一些思路。方法:选取SPF级Wistar雄性大鼠60只,随机数字表法将大鼠分成4组,模型组、正常组、阳性对照组、消渴汤组,除正常组外,其他3组大鼠制备糖尿病模型,造模后阳性对照组灌胃0.25g/kg的二甲双胍药液,消渴汤组灌胃35g/kg的消渴汤药液,模型组和正常组灌胃等剂量生理盐水,1次/d,连续灌胃6周。观察大鼠一般状况,末次给药后检测大鼠体质量、24h尿量情况,取大鼠腹主动脉血清5ml,ELISA法检测血清MCP-1、FINS、IL-6及TNF-α含量状况,全自动生化分析仪检测LDL-C、FBG、TG、HDL-C及TC含量状况,并计算HOMA-RI指数。结果:和正常组相比,模型大鼠体质量下降,24h尿量、HOMA-RI、FINS及FBG水平上升;和模型组相比,阳性对照组、消渴汤组大鼠体质量上升,24h尿量、HOMA-RI、FINS及FBG水平下降,差异均有统计学意义(P<0.05);和正常组相比,模型组大鼠血清LDL-C、TC、TG含量上升,HDL-C含量下降;和模型组相比,阳性对照组、消渴汤组大鼠血清LDL-C、TC、TG含量下降,HDL-C含量上升,差异均有统计学意义(P<0.05);和正常组相比,模型组大鼠血清MCP-1、IL-6及TNF-α含量上升;和模型组相比,阳性对照组、消渴汤组大鼠血清MCP-1、IL-6及TNF-α含量下降,差异有统计学意义(P<0.05)。结论:消渴汤可显著改善糖尿病大鼠胰岛素抵抗状况,降低血清炎症因子、血脂与血糖含量。
Objective:To analyze the effect of Xiaoke Decoction on liver glucose and lipid metabolism and inflammatory response for diabetic rats,and provide some ideas for the treatment of clinical patients.Methods:Sixty Wistar male rats of SPF grade were randomly divided into four groups:model group,normal group,positive controlled group and Xiaoke Decoction group.In addition to the normal group,the other three groups of rats were prepared for diabetes.After the model was established,the positive controlled group was given 0.25 g/kg of metformin solution,the Xiaoke Decoction group was given 35 g/kg of Xiaoke Decoction solution,the model group and the normal group were given the same dose of normal saline once a day,continuous gavage for 6 weeks.The general condition of the rats was observed.The body weight and 24 h urine volume were measured after the last administration.The serum of the abdominal aorta was taken 5 ml.The serum levels of MCP-1,FINS,IL-6 and TNF-αwere detected by ELISA.The automatic biochemical analyzer was used to detect the LDL-C,FBG,TG,HDL-C and TC content,and the HOMA-RI index was calculated.Results:Compared with the normal group,the body weight of the model rats decreased,and the 24 h urine volume,HOMA-RI,FINS and FBG levels increased.Compared with the model group,the body weight of the positive controlled group and Xiaoke Decoction group increased,24 h urine output,HOMA-RI,FINS and FBG levels decreased,the difference was statistically significant(P<0.05).Compared with the normal group,the serum LDL-C,TC,TG levels and HDL-C content in the model group were increased.Compared with the model group,the serum levels of LDL-C,TC and TG in the positive controlled group and Xiaoke Decoction group decreased,and the HDL-C content increased,the difference was statistically significant(P<0.05).Compared with the model group,the serum levels of MCP-1,IL-6 and TNF-αin the model group were higher than those in the model group.The content decreased,and the difference was statistically significant(P<0.05).Conclusion:X
