卡托普利作用不同时间点糖尿病合并慢性阻塞性肺疾病模型大鼠血清中IL-8和TNF-α变化趋势研究 被引量:3

摘要 目的研究卡托普利干预后不同时间点DM合并COPD模型大鼠血清中IL-8和TNF-α变化趋势。方法建立DM合并COPD模型,并分为Control组(正常大鼠,等量生理盐水灌胃)、合并模型组(模型大鼠,等量生理盐水灌胃)、Captopril治疗组(模型大鼠,5 mg/kg/d Captopril灌胃,连续6 w),大鼠ELISA试剂盒检测血清和肺组织IL-8和TNF-α含量;HE染色观察肺组织病理学变化,并进行评分。结果 (1)治疗1-6 w后,与合并模型组相比,Captopril治疗组血清中IL-8和TNF-α含量均显著降低(P均<0.05);(2)仅Captopril治疗组IL-8和TNF-α的水平与时间显著相关,随时间增加而逐渐降低(r均<0,P均<0.05);(3)治疗6 w后,与对照组相比,合并模型组肺组织中IL-8和TNF-α含量和病理学评分以及FPG值均显著增加(P<0.05),经Captopril治疗后,大鼠以上指标均显著降低(P<0.05);(4)肺组织和血清中IL-8和TNF-α的变化均呈显著正相关(P均<0.05)。结论经卡托普利治疗后,DM合并COPD模型大鼠血清及肺组织IL-8和TNF-α含量降低,血糖水平以及肺组织病理状况改善,且其作用效果随时间延长而增加。 Objective To investigate the changes of IL-8 and TNF-αin serum of DM combined with COPD model rats at different time points after captopril intervention.Methods DM combined with COPD model were established and then divided into 3 groups,which were the control group(normal rats,equal saline administration),the model group(DM+COPD model rats with equal volume of normal saline)and the Captopril group(DM+COPD model rats,intragastric administration with 5 mg/kg/d captopril solution for 6 weeks).The rat ELISA kit was used to detect the IL-8 and TNF-αlevels in rat serum and lung tissues.HE staining was used to observe the pathological conditions of the lung tissues,and pathological scores were obtained.Results(1)Compared with the combined model group after 1 to 6 weeks of treatment,the serum IL-8 and TNF-αlevels in the captopril treatment group were significantly reduced(both P<0.05).(2)The levels of IL-8 and TNF-αof the captopril treatment group were significantly correlated with time,and gradually decreased with time(both r<0,P<0.05).(3)Compared with the control group after 6 weeks of treatment,the IL-8 and TNF-αlevels,pathological scores and FPG values in the lung tissue of the combined model group were significantly increased(P<0.05).The above indexes of rats were significantly reduced(P<0.05).(4)The changes of IL-8 and TNF-αin lung tissue and serum were significantly positively correlated(both P<0.05).Conclusion After captopril treatment,the levels IL-8 and TNF-αin serum and lungtissue of DM combined with COPD model rats decrease,blood glucose levels and pathological conditions of lung tissue improve,and its effect increases with time.
