脑出血大鼠脑组织神经肽Y水平的变化(英文) 被引量:7

摘要 目的动态观测脑出血过程中大鼠血肿周边、下丘脑及脑干NPY含量的变化,探讨脑出血过程中中枢神经系统神经肽Y(NPY)含量的动态变化及意义。方法采用胶原酶和肝素联合注入尾状核的方法建立大鼠脑出血模型,测定出血前、出血后30min,6,12,24,48及72h血肿周边、下丘脑及脑干NPY的活性。NPY活性的测定采用放射免疫法。结果脑出血后血肿周边、下丘脑及脑干NPY的含量同步升高,并于24h达峰值,分别为犤(4.37±0.14),(9.06±0.15),(7.22±0.26)μg/gprotein犦,48~72h开始回落。但仍显著高于出血前(t=31.3216,12.5715,11.6487,P<0.01)。结论脑出血过程中,脑组织NPY活性增强,导致局部脑血流量减少,最终加重神经细胞损害。 Aim To observe dynamic changes of neuropeptide(NPY) level in periphery of hematoma,hypothalamus and brain stem of rats during cerebral hemorrhage and its significance.Methods Rat model of cerebral hemorrhage was established by combined injection of collagenase and hepatin into caudate nucleus and NPY activities in periphery of hematoma,hypothalamus and brain stem of rats before and at 30 minutes,6,12,24,48 and 72 h after hemorrhage were detected.Radioimmunity was used for detection of NPY activities.Results NPY activities in periphery of hematoma,hypothalamus and brain stem of rats increased sychronously each other,reached the peak at 24 h[(4.37± 0.14),(9.06± 0.15),(7.22± 0.26)μ g/g] and began decreasing from 48 to 72 h.But they were still significantly higher than that before hemorrhage( t=31.321 6,12.571 5,11.648 7,P< 0.01) .Conclusion During cerebral hemorrhage,NPY activity in brain tissue increases.
