摘要
目的:采用立体定向注射法向大鼠尾状核内注射50μL自体动脉血,制作脑出血动物模型。方法:实验于2003-09/12在南方医科大学第一附属医院神经内科实验室进行。取SD大鼠7只,随机分为实验组5只和假手术组2只。实验组用肝素化50μL微量注射器抽取50μL股动脉血,用立体定向仪将微量注射器针进针至尾状核中心,将血液分两次缓慢注入尾状核内(注射速度10μL/min)。假手术组完成立体定向术及股动脉抽血,但不注血。术后6h取脑进行病理观察,并进行图像分析。结果:所有实验组大鼠尾状核内均可见血肿形成。部分血液沿针道反流至同侧侧脑室及胼胝体,血肿平均容量(13.79±5.53)μL。假手术组未发现血肿。结论:该脑出血模型成本低,操作简单,与临床脑出血相似。
AIM: To establish intracerebral hemorrhage model in rats by injecting autologous arterial blood into caudate nucleus with double injection method. METHODS: The study was finished in the Department of Neurology,First Affiliated Hospital of Nanfang Medical University from September to December 2003.Seven SD rats were randomly divided into experiment group(5 rats) and control group(2 rats).50 μL blood was drawn from the femoral artery with a 50 μL heparinized injector.After the needle of the injector was placed into left caudate nucleus by stereotactic frame, blood divided into two parts was injected slowly(10 μL/min) into caudate nucleus,The control group underwent stereotactic operation and withdrawal instead of infusion of blood. Pathology was observed by extracting cerebral constitution 6 hours after operation and at the same time the image was analyzed. RESULTS:Each rat of experiment group showed a hematoma in caudate nucleus with flooding of blood by the way back to lateral ventricle and mesolobus of the same side alone the needle track.The average volume of hematoma was(13.79±5.53)μL.The hematoma was not found in control group. CONCLUSION:The intracerebral hemorrhage model is cheap, easy to operate and similar to the clinical intracerebral hemorrhage.
