摘要
目的探讨凉血化化瘀法治疗脑出血大鼠的可能机制。方法将60只Wistar大鼠以随机表法随机均分为空白组、假手术组、模型组、治疗1组、治疗2组和治疗3组,后四组采用两次注射自体动脉血法构建大鼠脑出血模型,术后3h治疗1、2和3组分别给予凉血化瘀合剂2.4、4.8和7.2mg·kg-1·d-1灌胃,其他三组给予等量生理盐水。应用放射免疫法测定血ET和TNF-a含量。结果与空白组比较,假手术组血ET和TNF-α轻度升高;与假手术组比较,模型组血ET和TNF-α明显升高;与模型组比较,治疗组血ET和TNF-α明显下降;治疗组间比较血ET和TNF-α下降相近。结论凉血化瘀法治疗大鼠脑出血后对神经功能具有保护作用,其机制可能抑制ET表达和TNF-α毒性有关。
Objective:To study the possible mechanism of Liangxuehuayu Mixture(凉血化瘀合剂 ), the Drug of TCM Remoring Heat Blood and Removing Blood Stasis,in the treatment of the rats of intracerebral hemorrhage.Methos:90 Wistar rats were randomized into 6 groups:the normal group,the sham group,the model group,the treatment group 1.the treatment group 2 and the treatment group 3.The rats in the latter four groups were subjected to intracerebral hemorrhage models by injecting autologous arterial blood into caudate nucleus with double- injection method.Three hours after modeling,treatment group 1 were given Liangxuehuayu Mixture the dosage of 2.4mg/(kg· d),treatment group 2 given 4.8 mg/(kg· d),and treatment group 3 given 7.2 mg/(kg· d).Treatment groups were given consecutive 7 days.The other 3 groups were given normal saline at the dosage of 2.4 mg/(kg· d).The contents of blood ET and TNF- α were detected by radioimmunoassay.Results:In comparison with normal group,the contents of ET and TNF- α in sham group rose lightly;in comparison with sham group,the contents of ET and TNF- α in model group rose remarkably;incomparison with model group,the contents of ET and TNF- α in treatment groups fell remarkably;among the treatment groups,the contents of ET and TNF- α was almost equally fallen.Conclusion:The Therapy of TCM Removing Heat Blood and Removing Blood Stasis in the treatment after intracerebral hemorrhage can protect nerve functions possibly by suppressing ET and TNF- α .
